Objective:To compare the effect of combination of tamoxifen and doxorubicin with the respective effect of tamoxifen (TAM) or doxorubicin (ADM) on human osteosarcoma cell MG-63.Methods:The mRNA expression of estrogen receptor (ER) was detected in MG-63through (reverse transcription PCR) RT-PCR. The morphological changes of cell were observed with inverted microscope and the cell viability was tested by MTT colorimetric assay, after TAM and ADM were added together or alone.Results:The expression of ERaand ERβwere found expressing in MG-63. The typical apoptotic change of cells was observed in drug treated groups, which was not occurred in the control group. MTT colorimetric analysis demonstrated that the inhibition rate of cell proliferation of the combined drug group was significantly higher than that of other groups (P<0.05).Conclusions:The expression of ER-aand ER-βcould be found in human osteosarcoma cell MG-63. Both TAM and ADM can inhibit the proliferation of the cell. When TAM is combined with ADM, the inhibition effect is significantly enhanced. One of the possible mechanisms might be that TAM enhanced sensitivity of cells to ADM.
