Study Of Salmon Calcitonin Influences RANK/RANKL/OPG Osteolysis Pathway Of Macrophage Induced By Wear Debris | | Posted on:2014-02-10 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Leng | Full Text:PDF | | GTID:2254330425972959 | Subject:Clinical Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective:To investigate the influence of salmon calcitonin on RANK/RANKL/OPG osteolysis pathway by detecting the variation of RANKã€RANKLã€OPG mRNA expressed by macrophage induced by wear debris with or without salmon calcitonin.Methods:We extracted polyethylene wear debris from interface membranes around loosened prostheses of a patient who ought to undergoing revision total hip arthroplasty.Wear debris with diameter in0.2um-10um were collected after filtrating.To confirm the best responding macrophage cell concentration by MTT method.According to the different concentration of salmon calcitonin interfering with macrophage induced by wear debris,we set up5groups randomly:Group A(only macrophage);Group B(macrophage induced by wear debris); Group C(macrophage induced by wear debris,with high concentration of salmon calcitonin intervention10-8mol/L); Group D(macrophage induced by wear debris,with middle concentration of salmon calcitonin intervention10-10mol/L);Group E(macrophage induced by wear debris,with low concentration of salmon calcitonin intervention10-12mol/L).We detected the variation of RANKã€RANKLã€OPG mRNA expressed by macrophage after cultivating for48hours.Results:1.The expression of RANK:the expression of RANK in group B is higher than it in other groups(P<0.01). The expression of RANK in group C is higher than it in group D and E(P<0.01),but there is no statistical differences between group D and E(P>0.05). 2.The expression of RANKL:the expression of RANKL in group B is higher than it in group A(P<0.05) and group Cã€Dã€E(P<0.01). The expression of RANKL in group D is different between group C and E(P<0.01).There is no statistical differences between group C and E(P>0.05).3.The expression of OPG:the expression of OPG in group A is higher than it in other groups(P<0.01).The expression of OPG in group B is lower than it in group C and D(P<0.05).There is no statistical differences between group B and E(P>0.05).There is statistical differences between group E and C,Also between group E and D(P<0.05).There is no statistical differences between group C and D(P>0.05).4. RANKL/OPG:the ratio of RANKL/OPG in group B is higher than it in other groups.After the influences of calcitonin, the ratio of RANKL/OPG falls down,and the ratio of RANKL/OPG in group C is the lowest.Conclusions:1.Macrophage induced by wear debris could make the expression of RANK,RANKL and OPG;2.The ratio of RANKL/OPG had shown a upward trend after stimulated by wear debris,which may induce the transformation from macrophage to osteoclasts;3.
