Purpose:By observe the influence of different concentrations catecholamines on the activity of LPS-induced HUVEC, the content of the cell supernatant TNF-a and the expression of VE-cadherin, to elucidate the effect of catecholamines on LPS induced HUVEC.Methods:1) Select the voluntarily donated umbilical cord of newborn infant of full-term pregnancy from healthy mothers, about25-30cm; isolate human umbilical vein endothelial cells, then cultivate them to generate new ones, and then frozen them, getting the third generation cells to be used in the experiment.2) Performed on LPS-induced human umbilical vein endothelial cells, to detect different concentrations of catecholamines in human umbilical vein endothelial cell activity using MTT method.3) Performed on LPS-induced human umbilical vein endothelial cells, to detect the content of cell supernatant TNF-a using enzyme-linked immunoadsordent assay (ELISA).4) Performed on LPS-induced human umbilical vein endothelial cells, to detect the expression of HUVEC cytoskeletal proteins (VE-Cadherin) using Western Blot method.Results:1)After the stimulation of LPS, the activity of HUVEC was significantly suppressed and with time, HUVEC activity was significantly decreased;2)Normally, the influence to the activity of HUVEC, under DA (1-10μM) and NE/E (0.1-1μM) is not significant. But with the increase of the drug concentration, the cell activity decreases significantly, the content of TNF-a increases, and the expression of VE-cadherin decreases.3)LPS group:DA (1-100μM) and NE/E (0.1-10μM) can antilipid the damage function of LPS-induced cell. But with the increase of concentration, the cell activity decreased,the content of TNF-a increased, and the expression of VE-cadherin decreased.Conclusions:l)The experiment confirmed the inhibit the activity of LPS to HUVEC;2). catecholamines (DA:1-100μM或NE/E:0.1-10μM) protective LPS-induced human umbilical vein endothelial cells injure;3.The protective effect of catecholamines (DA:1-100μM或NE/E:0.1-10μM) on LPS-induced Human umbilical vein endothelial cells, through Inhibition of TNF-a secretion and the expression of VE-cadherin increases.4.the catecholamines (DA>100μM或NE/E>10uM) produce toxic effects of drugs on HUVEC.
