To investigate the effect of magnesium ions on calcification in RVSMC induced by high calcium and high phosphorus culture medium,confirm whether magnesium reduce calcification in RVSMC via the inhibition of transdifferentiation of RVSMC into osteoblast-like cells and apoptosis.Method:RVSMC were cultured in vitro, adjusting medium concentration of calcium (Ca), phosphate (Pi) and magnesium ions with CaCl2,NaH2PO4and MgSO4. At different time points,the contents of calcium in cells were dected, cbfa-1and a-SMA mRNA was detected by Real Time PCR, apoptosis of RVSMC was detected by MTTResult:1.Compared with normal calcium and phosphorus group(control),Calcium content in the cells which cultured with high calcium or high phosphorus medium were increased significantly on7,10,14days.,the difference was statistically significant (P<0.01).On the14th day,compared with control,expression of cbfa-1mRNA increased significantly(P<0.05),but expression of a-SMA mRNA decreased significantly(P<0.05), apoptosis rate of RVSMC increased significantly (P<0.01).2.We cultivated RVSMC with different concentrations of magnesium ions(1.0mmol/l,2.0mmol/l,3.0mmol/l),calcium (3.0mmol/l) or phosphorus (2.4mmol/l) for7,14days.The contents of calcium in cells fell with the magnesium ions concentration in culture medium, a dose dependent(P<0.05).3.We cultivated RVSMC with different concentrations of magnesium ions (1.0mmol/l,2.0mmo1/1,3.0mmol/l),calcium (3.0mmol/l) and phosphorus (2.4mmol/l) for14days.Compared with high calcium and high phosphorus cultrue,the expression of a-SMA and cbfa-1mRNA of high magnesium group was significantly different (P<0.05).4. We cultivated RVSMC with different concentrations of magnesium ions(1.0mmol/l,2.0mmol/l,3.0mmol/l),calcium (3.0mmol/l) and phosphorus (2.4mmol/l) for7days.Apoptosis rate of RVSMC fell with the magnesium concentration in culture medium, a dose dependent (P<0.05).Conclusion:1.It indicates that high calcium or high phosphorus may increase calcification in RVSMC,via promoting them to the osteogenesis transdifferentiation and apoptosis.2.High magnesium can reduce calcification in RVSMC induced by high calcium and high phosphorus culture medium, via the inhibition of transdifferentiation of RVSMC into osteoblast-like cells and apoptosis dose-and time-dependent manners totally.
