Objectives:To study the level of profilin-1, receptor for advanced glycation end products (RAGE) in the coronary artery disease (CAD) combined with type2Diabetes Mellitus (DM) and the correlation and clinical meaning between profilin-1, RAGE and the Gensini score system for the degree of coronary artery stenosis.Methods:89people from August2012to February2013in the Department of Cardiology, and Medical Examination Center, Xiangya Hospital, Central South University were enrolled in this study. Cases in3different groups are as:1) normal control group (Con):N=27;2) CAD group:N=31;3) CAD combined type2DM (CAD+DM):N=31. All the CAD, CAD combined with type2DM patients and part of normal people were received Coronary Angiograph (CAG). The liver function, renal function, plasma fasting blood glucose, glycosylated hemoglobin, glycated serum protein, lipids, high-sensitivity C-reactive protein were all checked after collecting2ml after8h overnight fasting blood samples, by automatic biochemical analyzer. Plasma profilin-1and RAGE were checked by enzyme-linked immunosorbent assay (ELISA). All the degree of coronary stenosis were calculated according to the Gensini scoring system.Results:1. Plasma profilin-1level was significantly higher in CAD, CAD+DM than in Con group (p<0.01), and plasma profilin-1level of CAD+DM was significantly higher than CAD group (p<0.01);2. Plasma RAGE level was significantly higher in CAD, CAD+DM than in Con group (p<0.01), and plasma RAGE level of CAD+DM was significantly higher than CAD group (p<0.01);3.The degree of Gensini score was significantly higher in CAD, CAD+DM than in Con group (p<0.01), and the degree of CAD+DM was significantly higher than CAD group (p<0.01);4.The plasma profilin-1level and Gensini scores was a positive correlation (R2=0.7494;p<0.01); plasma RAGE level and Gesini scores was a positive correlation (R2=0.8424;p<0.01); plasma profilin-1and RAGE level was positively correlated (R2=0.7161;p<0.01).Conclusions:1. The plasma profilin-1and RAGE level in CAD+DM were significantly higher than CAD;2. The plasma profilin-1and RAGE was a positive correlation;3. The plasma profilin-1, RAGE and Gensini scores were positively correlated, suggests that the plasma profilin-1and RAGE level may reflect coronary artery stenosis to some extent. Objective:To explore the effect advanced glycation end products (AGEs) on the proliferation and expression of profilin-1in rat vascular smooth muscle cells (RVSMCs).Methods:RVSMCs were cultured in vivo. AGEs stimulated RVSMCs for24h in a different final concentration among50μg/ml,100μg/ml and150μg/ml, MTT assay was used to observe the proliferation of RVSMCs. AGEs stimulated RVSMCs for24h in a final concentration of150μg/ml, Real-Time PCR method was used to detect the expression of the RAGE and profilin-1mRNA.Results:1. All the concentration of AGEs can induce the proliferation of RVSMCs (p<0.05),and the proliferation of RVSMCs in the final concerntration of150μg/ml was significantly higher than other group (p<0.05);2. AGEs stimulated RVSMCs for24h in a final concentration of150μg/ml, the expression of profilin-1and RAGE mRNA were significantly higher than the control group (p<0.05).Conclusion:AGEs can up-regulate the expression of RAGE and profilin-1mRNA in RVSMCs, suggested that AGEs may induce the expression of profilin-1in RVSMCs through the RAGE, promote the proliferation of RVSMCs.
