| Aim:To observe the expression of HSP90in serum and thoracic aorta of diabetic rats and analysis the correlations between HSP90and oxidative stress.To assess the effects of probucol on the expression of HSP90of diabetic rats.Methods:50male8-week-old SD rats were randomly divided into normal control group:N group (n=10) and high fat diet group:G group (n=40). Rats of group G were intraperitoneally injected by STZ(30mg/kg) while rats of group N injected by Citric acid buffer after4weeks.Random blood glucose≥16.7mmol/L and maintain2weeks as successful models. The diabetes rats were divided into two groups:adiabetes control group(DM group),probucol treatment group(PB group). PB group rats were treated with probucol(500mg/kg.d) by intragastric administration,N group and DM group were treated with CMC.8weeks later, we measured serum triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol(HDL), low density lipoprotein cholesterol(LDL). The expression of MDA, SOD, HSP90in serum was assessed by ELISA. The degree of injury of the thoracic aorta was observed by optical microscope. HSP90and eNOS expression were determined by immunohistochemistry method.Results:(1) The level of TC and HDL of PB group rats was lower than that in DM group rats(P<0.01),while there was no difference in the level of TG and LDL(P>0.05).And the level of TG,TC,LDL in DM group rats was higher than that in N group rats(P<0.05). There was no difference in the level of HDL in two group rats(P>0.05).(2) The expression of superoxide dismutase(SOD)in serum in DM rats was significantly lower than N rats (P<0.001)and the expression of HSP90(P<0.05)and malonaldehyde(MDA)(P<0.001)in DM rats was significantly higher than N rats.The expression of SOD in group PB rats was higher(P<0.01),HSP90(P<0.001)and MDA was lower(}<0.001)than group DM rats. The expression of HSP90and SOD were negative correlated(r=-0.817,P<0.001). The expression of HSP90and MDA were positively correlated(r=0.658,P<0.001).(3) The endothelial cells of thoracic aorta of DM group rats were widely shedding and the smooth muscle cell were hyperplasia and irregular. The endothelial cells of thoracic aorta of PB group rats were partly shedding and the smooth muscle cell were slightly hyperplasia and irregular. However, The endothelial cells and smooth muscle cell of thoracic aorta of N group rats were smooth and tidy.(4) The expression of HSP90in thoracic aortic was significantly increased in DM group rats compared to N group rats (P<0.05). While there is no difference in the expression of HSP90in thoracic aortic in PB group and DM group rats(P>0.05). However, There is no difference in the expression of eNOS in thoracic aortic(P>0.05).Conclusions:(1)The expression of HSP90both in serum and thoracic aortic was increased in diabetic rats, and was positively correlated with the oxidative stress.(2)HSP90in serum of diabetic rats can be reduced by treating with Probucol. Probucol have a protective effect on diabetic macroangiopathy by reducing the oxidative stress. |
