ObjectiveTo investigate the expression of Girdin in esophageal squamous cell carcinoma (ESCC) tissues.Explore the relationship between the expression of Girdin and pathological factors,progosis of ESCC.Meantime,with the siRNA against Girdin transfected ECA109cell lines to inhibit the expression of Girdin,investigated the cell proliferation and invasion after transfection.Methods1.Immunohistochemistry method was used to detect the expression of Girdin in94ESCC tissues and78corresponding noncancerous tissues.The relationship of the expression of Girdin and pathological factors, progosis in ESCC were subjected to statistics analysis.2.We selected blank group,the effective siRNA group and negative control group to following experiment.Girdin-specific siRNA was transfected ECA109cell,the expression levels of Girdin mRNA were determined by RT-PCR.3.Testing the cell proliferation after transfection using MTT.4.Determining invasion by transwell experiment.Results1、The result of immunohistochemistry indicated Girdin were high expression in esophageal tissues, In contrast,none of the corresponding noncancerous tissues showed a significant level of immunohistochemical staining for Girdin (P=0.000).There were no relationship between the expression of Girdin with age,sex and tumor location.However,Girdin overexpression was significantly associated with T stage(P=0.001), regional lymph node involvement(P=0.022) and tumor node metastasis(TNM)stage(P=0.004).The patient with Girdin-positive had poorer5-year survival rate than those with Girdin-negative (P=0.000).Univariate analysis showed that the5-year survival rate of ESCC was correlated with T stage,lymph node metastasis,TNM stage and high expression of Girdin(P<0.01).Multivariate analysis including the above four parameters identified TNM stage and the expression of Girdin as independent influence factors of the5-year survival rate.2.After24hours of transfection,RT-PCR showed that after transfecting siRNA into ECA109cell,the Girdin mRNA inhibition rate was76.52%.Compared with other groups,the difference was statistically significant.3.MTT showed that the inhibition ratio of cell proliferation were26,74%,37.24%,28.47%in24h,48h,72h respectively after trans fection.The result was statistically significant (P<0.01).4.Invasion assay showed that the number of cells which crossed the membrane was (0.072±0.005) in siRNA transfection group,(0.260±0.012) in blank group and (0.238±0.009) in negative control group (P<0.01).Conclusions1.The expression of Girdin was higher in esophageal carcinoma than that of the corresponding noncancerous tissues. The expression of Girdin is associated with T stage, lymph node metastasis and TNM stage.Girdin may be a prognostic marker in ESCC.2.Inhibition the expression of Girdin could significantly reduce the cell proliferation and invasion on esophageal carcinoma ECA109cell line.
