The Relationship Between P-gp Expression And Neoadjuvant Chemotherapy In Breast Cancer

PurposeThis study is to investigate the expression pattern of P-gp in breast cancer and evaluate the relationship of its expression with neoadjuvant chemotherapy, which would provide the clinicians a basis for indivi-dualized treatment for patients..MethodFrom November2011to January2012,64female patients with primary breast cancer from the breast surgery department in Xiangya Hospital were selected for study. They were randomly assigned to receive3-4courses of treatment of either anthracycline+cyclopho-sphamide, paclitaxel, cyclophosphamide, paclitaxel class+cyclophos-phamide combination neoadjuvant anthracycline chemotherapy. Immu-nohistochemistry was used to determine the expression level of P-gp in the breast cancer tissue obtained by hollow puncture before and after the chemotherapy. The samples collected before therapy were used to evaluate the expression patten of P-gp in breast cancer while the tissue samples obtained after treatment were used to examine the influence of neoadjuvant chemotherapy on P-gp expression. The SPSS17.0system was used for statistical analysis. Result39people out of the64analyzed patients showed positive expression of P-gp. The positive expression rate is60.93%.Also, the study demonstrates that there is no correlation of P-gp expression in breast cancer with the biological indicators of breast cancer, including patient age, tumor size, lymph node metastasis stage, ER and PR status and menopause status.Moreover, it was found in the study that the expression of P-gp and protein level of HER2and P53are positively correlated in the breast cancer tissues of the patients evaluated.P-gp express negative patients, chemotherapy better curative effect。 Cyclophosphamide merger purple sweater class or merge anthrax-cycline-based chemotherapy scheme can make the P-gp positive expression down. TAC chemotherapy which is the regimen of chemo-therapy to after the effects of P-gp decline no better than AC or TC scheme.ConclusionP-gp expression in breast cancer is not associated with the patient’s age, tumor size, lymph node metastasis, ER, PR, status or menopause status. However, its expression is positively correlated with the expres- sion of HER2and P53.
