Objectives(1) To evaluate the effect of local heating technique to the serum insulin,Peak time(Tmax),Peak concentration(Cmax)and insulin AUC while subcutaneous insulin injection.(2)To evaluate the effect of local heating technique to the concentration regulation of blood glucose while subcutaneous insulin injection.MethodsTwenty-four heathy Newsland white rabbits,weighing2.0-3.Okg, male,were used ALX solution to successfully established T1DM model via ear vein.After that,they were randomly divided into control group and experimental group.The diabetic rabbits of experimental group(the normal rectal temperature is38.5-40℃) used the heat patch to heat the infusion site to41-43℃for10minutes before injecting insulin and lasting30minutes after injecting insulin; the diabetic rabbits of control group didn’t do anything at the infusion site.Venous blood samples for serum insulin and blood glucose measurements were drawn at-10、0、5、10、15、25、30、40、50、60、70、90、120、180and240min with2ml.Considering the frequence of drawing blood was too many,we supplied0.9%NaCl for diabetic rabbits with10ml via ear vein to avoid hemorrhagic shock at0,30,60and180min.Use SPSS17.0to analyse the effect of the absorption of insulin and blood glucose with heat patch and without heat patch.Results(1)5%ALX solution was injected into Newsland white rabbits via ear vein to establish T1DM model.The success rate was83.3%and death rate was8.3%.(2) The abdomen temperature of experimental group was higher than control group(P<0.05).(3) Comparison of control group and experimental group:the experimental group was associated with significantly decreases in Tmax、 and increases in Cmax、insulinAUCo-30、insulinAUCo-60and insulinAUCo-90.(all, P<0.05)(4) The serum insulin with heat patch was significantly higher than without heat patch,F=15.4,P<0.05.(5) Comparison of the decrease of blood glucose of control group and experimental group:they were significantly different at40,90,120and240minutes(all,P<0.05), the experimental group was better decreased blood glucose.Conclusions(1)ALX was injected for three times can successfully establish T1DM model.(2)Heating the insulin site can decrease Tmax and increase Cmax,all this can accelerate insulin absorption.(3)Heating the insulin site can more quickly decrease blood glucose,to avoid high blood glucose for a long time and reducing blood glucose excursion.
