The Influence Of AEoTGE On The Expression Of Keloid Fibroblast Ⅰ Collagen

Purpose: Discussing the influence of AEoTGE on the expression of Keloidfibroblast Ⅰ collagen.Method:1.Fibroblasts cultured in vitro with tissue block method.2.Observing thecell morphology with inverted microscope. Identifying the cell ultrastructure withTransmission Electron Microscope.3. Observing the inhibition of fibroblast with CellCounting Kit8after medicating different concentrations of AEoTGE（7.5，10，15，25，50，75，100g/L）24hours later. Calculating the median inhibition concentration andFinding out a drug concentration which have no significant difference with controlgroup.4. Checking the expression of Ⅰ collagen with Human Ⅰ collagen Elisa kit.Result:1.10Keloid tissue from the patient all longed out of the cell which canbe used in this research.2.With the increasing of concentration, AEoTGE have a moreand more strong inhibitory effect on the fibroblast. It have the significant differencewith control group（p<0.05）.3.The median inhibition concentration is35g/L, the ODvalue of medicine group of10g/L have no significant difference with control group（p﹥0.1).4.The expression of Ⅰ collagen have significantly reduced with Human Ⅰcollagen Elisa kit after10g/L AEoTGE added. It have the significant difference withcontrol group and it have the statistical significance（p<0.01）.Conclusion: After the action of AEoTGE, the expression ofⅠcollagen ofhuman Keloid fibroblast cultured in vitro have significantly reduced, it have thestatistical significance.
