Olaquindox is a kind of antiseptics widely used in livestock breeding in chinaalthough it is forbidden in European Union. There was many researches focused on thetoxicity of olaquindox in Liver and kidney. But it is not clear whether olaquindox alsohas toxic effect on pancreatic islet cells. In this study, the genotoxic potential ofOlaquindox was investigated in pancreatic islet B cells of the DM rat(INS-1) in cultureusing single cell gel electrophoresis (SCGE) assay. We observed the expression of p53protein exposed to olaquindox24h and48h using western blot technique and in thisway we can explore the increased expressions of oncogene may relate to the persistentdamage of DNA. To further the reaction mechanisms we monitored the level ofreactive oxygen species (ROS) production with the2,7-dichlorofluorescein diacetateassay and measured the changes of lysosomal membrane stability with Acridineorange(AO). Olaquindox at200-1600ug caused the increase of the DNA migration,the expressions of p53, the levels of intracellular ROS and the repture of lysosomal.Accordingly, we can conclude olaquindox exerts genotoxic effects on pancreatic islet-βcells of the DM rat, probably through oxidative DNA damage induced byintracellular ROS and repture of lysosomal. Besides that, the increased expressions ofoncogene may relate to the persistent damage of DNA.
