Objective:Diseases which related to lumbar intervertebral disc degeneration seriously affect life of patients and carry heavy social burden, but the mechanism of lumbar intervertebral disc degeneration is still unclear."Bu Shen Huo Xue Shu Jin" decoction has great curative effect in the treatment of diseases which related to lumbar intervertebral disc degeneration, and its mechanism need to further research.Methods:Fifteen Bordeaux goats are divided to three groups, thsoe are animal model feeded with "Bu Shen Huo Xue Shu Jin" decoction (group A), animal model feeded with normal saline (group B) and sham-operation group,(group C).Original dates of all lumbar intervertebral disc are detected by MR before operation. Full-endoscopic technique of minimally invasive surgery is used to make animal model of lumbar intervertebral disc degeneration. Sham-operation group use the same technique but the structure of lumbar intervertebral disc is not injured. After operation, animals in group A are feeded with extracting solution of "Bu Shen Huo Xue Shu Jin" decoction according to body surface area. Group B and group C are feeded with normal saline. After two months MRI test is taken again and lumbar intervertebral discs are harvested after animals sacrificed. Comparation of MRI scores, histology scores and gene expression including HIF-1、TNF-α、TGF-β、NFκB、IL-1in nucleus pulposus tissue are taken in all groups.Results:There is a statistically significant difference in MRI scores and histology scores of the three groups (P<0.05),except group A and group B in MRI. Gene expression also has a significant difference in all groups, and the gene expression of HIF-1has a statistically significant difference (P<0.01). The expression of TNF-a has a statistically difference between group A and group C, as well as group B and group C (P<0.05), but the difference between group A and group B have no statistically mean (P>0.05)Conclusions:Full-endoscopic technique of minimally invasive surgery successfully estabilish a goat model of lumbar intervertebral disc degeneration."Bu Shen Huo Xue Shu Jin" decoction inhibits the progress of lumbar intervertebral disc degeneration related to its function of inhibits inflammatory mediators expression and promotes protective factors expression in lumbar intervertebral disc tissue.
