Objective: Previous studies have found the P16plays an important role in manytumors. However, there have rarely about the expression of P16in humanconjunctival malignant melanoma and conjunctival nevi and their releationship withclinical stage and prognosis of conjunctival malignant melanoma. The purpose of thisstudy is to observe the expression of P16,Ki-67,HMB45in human conjunctivalmalignant melanoma and conjunctival nevi, and to explore the relationship betweenP16,Ki-67,HMB45and the clinical stage and prognosis of conjunctival malignantmelanoma.Methods:Retrospective study of20cases of malignant melanoma of conjunctivaby diagnosis at Zhongshan Ophthalmic Center between April2002and July2010.Wereview the patients’ clinical data, clinical and pathological features and performimmunohistochemical staining.20cases of conjunctival nevi as a control group.TNM stage according to the American Joint Committee on Cancer (AJCC)2010Cancer Staging Manual, statistical analysis of the relationship betweenP16,Ki-67,HMB45and clinical stagie, prognosis of conjunctival malignantmelanoma.Results: In conjunctival malignant melanoma, the negative expression rate ofP16is60%(12/20),low expression rae is20%(4/20),mediun-high expression rae is20%(4/20); the negative expression rate of Ki-67is20%(4/20),low expression rae is25%(5/20),mediun-high expression rae is55%(11/20); the negative expression rate ofHMB45is10%(2/20),low expression rae is15%(3/20),mediun-high expression rae is75%(15/20).In conjunctival nevi, the negative expression rate of P16is5%(1/20),lowexpression rae is20%(4/20),mediun-high expression rae is75%(15/20); the negativeexpression rate of Ki-67is75%(15/20),low expression rae is25%(5/20); the negativeexpression rate of HMB45is90%(18/20),low expression rae is10%(2/20). Theexpression of P16,Ki-67,HMB45between conjunctival malignant melanoma andconjunctival nevi exist statistical differences(P<0.05). The expression of P16andKi-67in conjunctival malignant melanoma was negatively correlated(r=﹣0.786, P<0.05). The P16expression was negatively correlated with the recurrence rate andclinical stage of conjunctival malignant melanoma(r=﹣0.618,P<0.05;r=﹣0.491,P<0.05). Ki-67expression was positively correlated with the recurrence rate ofconjunctival malignant melanoma(r=0.504,P<0.05). HMB45has no relationshipwith the recurrence rate and clinical stage of conjunctival malignant melanoma(X2=2.629,P>0.05;X2=0.317,P>0.05).Conclusions: P16expression in conjunctival nevi stonger than conjunctivalmalignant melanoma; HMB45, Ki-67expression in conjunctival malignant melanomastronger than conjunctival nevi. The expression of P16and Ki-67in conjunctivalmalignant melanoma was negatively correlated. The P16expression was negativelycorrelated with the recurrence rate and clinical stage of conjunctival malignantmelanoma; Ki-67expression was positively correlated with the recurrence rate ofconjunctival malignant melanoma. P16and Ki-67may be used as prognosticindicators of conjunctival malignant melanoma.
