Objective:This thesis aims to study the expressions of gene EGR-1and HPSE, to make apreliminary probe into these two genes’ function in the attack of human embryonalcarcinoma of testis and tries to analyze the link between the genes in the mutualexpression of the disease, which helps people learn about the biological behavior ofhuman embryonal carcinoma and provide support for bio-immuno therapy andpostoperative targeted therapy in the future.Methods:The thesis chooses five cases from the tissue of human embryonal carcinoma oftestis, six cases from the tissue of normal testis, studies the expressions ofEGR-1mRNA, HPSE mRNA and related protein, and analyzes the clinicalsignificance and correlation of gene EGR-1and HPSE in human embryonalcarcinoma of testis.Results:(1)EGR-1mRNA expressed in both normal and embryonal carcinoma oftesticular tissue.The differences in the optical density ratio between EGR-1belt andcorresponding referredβ-actin belt, and the comparison between normal andembryonal carcinoma of testicular tissues have no statistical significance.(ρ>0.05).(2)HPSE mRNA expressed in embryonal carcinoma of testicular tissue,while noexpression in normal tissue.(3)EGR-1protein expressed in both normal and embryonal carcinoma oftesticular tissues.The differences in the optical density ratio between EGR-1belt andcorresponding referred β-actin belt, and the comparison between normal andembryonal carcinoma of testicular tissues have statistical significance.(ρ <0.05)(4)HPSE protein expressed in embryonal carcinoma of testicular tissue,while noexpression in normal tissue.Conclusions:(1)The expression of gene HPSE in human embryonal carcinoma of testis may have a connection with occurrence, invasion, metastasis of human embryonalcarcinoma of testis.(2)Gene EGR-1may play an important role in the development of humanembryonal carcinoma of testis.(3)The mutual expression of gene EGR-1and HPSE in human embryonalcarcinoma of testis may have a synergistic action(EGR-1regulates HPSE),whichleads to early metastasis, high malignancy and poor prognosis of human embryonalcarcinoma of testis.Gene EGR-1and HPSE may offer a new choice tobio-immunotherapy and postoperative targeted therapy of human embryonalcarcinoma of testis.
