| Background and Objective:Normal epithelial cell specific-1is also known as a member of the human tissuekallikrein10(KLK10) family, which coded the Serine protease (hKs). The aim of thisstudy was to investigate colon cancer occurrence mechanism by explore theexpression of NES1and estrogen receptor in the normal colonic mucosa cells andcolon cancer cell lines.Material and Methods:Culture the normal colonic mucosa cells and colon cancer cell lines(HT29,HCT116) respectively, Inverted microscope to observe the morphologicalcharacteristics of cell growth.Isolated the RNA, quantitative PCR to detect theexpression of NES1and estrogen receptor in cells.Results:1. NES1expression in the normal colonic mucosa cells was significantly higherthan in the colon cancer cell lines.2. Estrogen receptor was expressed both in normal colonic mucosa cells andcolon cancer cell lines(HCT116and HT29); Estrogen receptor was significantlydown-regulated in colon cancer cell lines(HCT116and HT29cells).Conclusion:1. NES1gene was significantly higher expression in colon cancer cell lines, itcan be used as auxiliary markers for the diagnosis of colon tumors.2. The expression of NES1gene and estrogen receptor was negativelycorrelated. |
