Objective:1.To examine the expression of miR-145,Akt and p-Akt protein in Normal ovary,benign and malignant epithelial ovarian tumor tissues.2.To explore the roles of miR-145,Akt and p-Akt protein in the tumorigenesisand progression of human epithelial ovarian cancer, and the relationship amongmiR-145,Akt and p-Akt protein in the carcinogenesis of ovarian cancer。Methods:A total of10normal ovarian cases,19benign epithelial ovarian tumor cases and17epithelial ovarian cancer cases were collected from Maternal and Child HealthHospital of Jiangxi Province.The expression of miR-145was examined by semiquantitative RT-PCR,and Western blot was used to measure Akt and p-Aktexpression.meanwhile, we analyzed these data with statistical software.Results:1.The expression of miR-145was detected in normal ovarian tissue,benign andmalignant epithelial ovarian cancer tissues.and the expression level presented withsignificant differences (F=12.267,P<0.05).MiR-145expression in epithelial ovariancancer group was lower than that in other histologic groups (P<0.05).The expressionlevel of miR-145has no relation with FIGO stage and pathologic types(P>0.05).2.The Akt protein was expressed in normal ovarian tissue, benign and malignantepithelial ovarian cancer tissues,but presented with no significantdifference(F=1.588,P>0.05).3.The p-Akt protein was often detected in normal ovarian tissue, benign andmalignant epithelial ovarian cancer tissues.and the expression level presented withsignificant differences(F=17.525,P<0.05).p-Akt expression in epithelial ovariancancer group was higher than that in other histologic types(P<0.05).The expression level of p-Akt was related to the FIGO stage (P<0.05),but has no relation withpathologic types(P>0.05).4.There was a negative correlation between miR-145and p-Akt (r=-0.492,p=0.001).The expression between miR-145and Akt,Akt and p-Akt had nocorrelation(r=0.088,P=0.56;r=0.180,P=0.232).Conclusions:1.miR-145,Akt and p-Akt protein were expressed in normal ovarian tissue,benign and malignant epithelial ovarian cancer tissues.2.Low-expression of miR-145maybe associated with the tumorigenesis ofhuman epithelial ovarian cancer. High-expression of p-Akt might play certain roles inthe tumorigenesis and the FIGO stage of human epithelial ovarian cancer.3.Low-expression of miR-145and excessive activation of Akt have negativecorrelation in epithelial ovarian cancer.
