Objective: To observe the influence in the growth and proliferation of thetransfected gene XPD on hepatoma cell SMMC-7721,and the expression of XPD、Ets-1and Cdk6.Methods:We transfected pEGFP-N2-XPD into human SMMC-7721cells byLipofectamine2000TM. There were four groups in this study including SMMC-7721-pEGFP-N2-XPD group(XPD group), SMMC-7721-pEGFP-N2group(N2group),Lipofectamine group(Lip group) and blank control group. The expressions of XPD,Ets-1and Cdk6were detected by RT-PCR and Western blot. Flow cytometry(FCM)was used to analyze the cell cycle. The cell proliferation was measured by MTT.Results:In four groups of cells,the expression of XPD mRNA and protein wassignificantly increased in XPD group(P<0.01), the expression of Ets-1and Cdk6mRNA and protein were reduced obviously(P<0.01),they had no difference betweenthe other three groups. Flow cytometry showed that XPD prevented the hepatomacells from entering the S stage, stopping in the G1stage. The proliferation ability wasdistinctly inhibited after transfected by XPD gene.Conclusion: The wild-type XPD gene may regulate the SMMC-7721cellsproliferation by inhibiting the Ets-1and Cdk6gene.
