AIM:To build the miRNA495and miRNA551a of eukaryotic expression vectorstargeting PRL-3gene, and to observe the PRL-3gene expression in SGC7901gastric cancer cells after the vectors stably transfected SGC7901gastric cancer cells.METHODS:The specific pre-miRNA495and pre-miRNA551a sequencestargeting PRL-3gene were designed and synthesized.Then ligation with pcDNA6.2-GW/EmGFP-miR in order.The correct sequence were confirmed.The recombinantvector were transfected into SGC7901gastric cancer cells with the help ofLipofectamine2000, then the suppression effect targeting PRL-3was identified byreal-time PCR and western-blot test.RESULTS:The PRL-3pre-miRNA495and pre-miRNA551a recombinant plasmidwere successfully constructed, and it promoted the miRNA495and miRNA551aexpressions in SGC7901gastric cancer cells effectively, at the same time it inhibitedthe PRL-3mRNA and protein expressions in SGC7901gastric cancer cellseffectively.CONCLUSION:The miRNA495and miRNA551a expression vector for silencingPRL-3can be constructed successfully;which can effectively promote themiRNA495and miRNA551a expressions and significantly inhibit the expressions ofPRL-3mRNA and protein of SGC7901gastric cancer cells.
