Objective:The purpose of this study is to clarify the tea polyphenols has the antopxidationeffect to trabecular cells in primary open angle glaucoma. And provides theexperimental basis for the application of tea polyphenols in the Department ofophthalmologyMethods:1、material: recovery HTC, GTC and go down to the future generation to3-5.put into the DMEM application solution, cultured in5%CO237oC conditions.2、Experimental groups: divide the cells in the logarithmic growth phase intofour groups randomly: Control group, the experimental group1,2,3,add theconcentration of5,10,20mg/L TP into the experimental group of1,2,3respectively while adding the same amount of DMEM application into the controlgroup.After cultured for24hours, measure cytochrome C (Cytochrome C, CytC)release from trabecular cells by Western blot. Measure the expression amount ofcomplexes Ⅰ messenger RNA (Complex I Messenger RNA, Complex I mRNA) intrabecular cells by fluorescence quantitative polymerase chain reaction (Fluorescencequantitative PCR).And the apoptosis rate of trabecular cell is measured by flowcytometry.Result:1、The influence of TP on the growth of cultured human trabecular meshworkcellsCompared with POAG group, the apoptosis rate decrease in the experimentalgroup1,2,3, the difference between them is statistically significant (P<0.05); withthe concentration of tea polyphenols increase, the apoptosis rate in the experimentalgroup1,2,3decrease, which means the difference between them is statisticallysignificant (P<0.05)2、Measure the release amount of CytC from trabecular cells by Western blot Compared with POAG group, the release amount of CytC decrease in theexperimental group1,2,3, the difference between them is statistically significant(P<0.05); with the concentration of tea polyphenols increase, the release amountof CytC in the experimental group1,2,3decrease, which means the differencebetween them is statistically significant (P<0.05)3、Measure the expression amount of complexes Ⅰ messenger RNA (ComplexI mRNA) in trabecular cells by PCRCompared with POAG group, the expression amount of Complex I mRNAdecrease in the experimental group1,2,3, the difference between them is statisticallysignificant (P<0.05); with the concentration of tea polyphenols increase, theexpression amount of Complex I mRNAin the experimental group1,2,3decrease,which means the difference between them is statistically significant (P<0.05)Conclusion:Exogenous tea polyphenols can protect POAG patients trabecular cells onoxidative damage;In a certain range of concentration, while increasing the concentration of teapolyphenols, the antioxidant effect is enhanced.
