Expressions Of ERCC1and FANCD2and Clinical Outcomes Of Platinum-based Chemotherapy In Completely Resected NSCLC

Objectives:1、To investigate the correlation between ERCC1and FANCD2protein levelsand clinicopathological features in stage IIIA-N2non-small cell lung cancer（NSCLC）.2、To analyze the prognostic significance of ERCC1and FANCD2protein incompletely resected stage IIIA-N2NSCLC.Methods:Immunohistochemical analysis was used to evaluate the expressions of ERCC1and FANCD2in completely resected stage IIIA-N2NSCLC of60cases.Thecorrelation between the expressions of the markers and clinicopathological featuresand prognoses were analyzed. All the analyses were performed with the use of SPSS18.0.Results:1、 The expression of ERCC1and FACND2protein in different lungtissues.Among60cases,the mean level of ERCC1and FANCD2proteins expressionin tumor tissue specimens was51.7%（31/60）and48.3%（29/60）,repectibely.Among9cases of nontumor tissue, the expression of ERCC1and FANCD2protein was allnegative. The expression of ERCC1and FANCD2protein in tumor tissuesignificantly higher than in nontumor tissue (P=0.003, P=0.008).2、No significant correlation was found between expression of ERCC1andFANCD2protein and clinicopathological features in stage IIIA-N2NSCLC.3、The correlation between the expression of ERCC1and FANCD2protein inNSCLC. Their expressions which were all positive or negative were respectively24cases. The expression of ERCC1was positive with negative expression of FANCD2were7cases.The expressions of FANCD2were positive with negative expression of ERCC1were5cases. There was correlation between the expression of ERCC1andFANCD2protein in NSCLC（P=0.000,r=0.602）.4、The relationship between clinicopathological features,The overall survival ofother type is shorter significantly than squamous carcinoma and adenocarcinoma.（P=0.017）。Platinum-based chemotherapy significantly prolonged overall survivalwith ERCC1or FANCD2negative (P=0.019, P=0.001).Conclusions:1、The expression of ERCC1and FANCD2protein in tumor tissue significantlyhigher than in nontumor tissue.2、There was a positive correlation between the expression of ERCC1andFANCD2protein in NSCLC.,and may be a link in the process of DNA repair.3、FANCD2expression was independent prognostic factors in the patients.Lower expressions of ERCC1and FANCD2have better outcomes.
