Objective: To observe the effects of basil polysaccharide on weight,survival time and tumor volume of mice with H22hepatoma cells, and vieweffect of basil polysaccharide on the signaling pathway formed bylymphatic endothelial growth factor and its receptor VEGF-C/VEGFR-3through further observation of expression of podoplanin which is thelymphatic endothelial specific markers, to investigate the mechanism ofbasil polysaccharide’s anti-tumor lymphatic metastasis, and providetheoretical and experimental basis for the clinical application of basilpolysaccharide.Methods: Copy lymph node metastasis model of mouse with H22hepatomacells, the mice are divided to saline control group, mitomycin group,high-dose basil group, high-dose basil and mitomycin group, small-dosebasil and mitomycin group, to observe the life cycle and tumor volume ofthe mice, it’s a total of three batches of experiments; recopy lymphnode metastasis model of mouse with H22cells, to treat that with salinecontrol group, large, medium and small-dose basil group, detect effectson regional lymph node tumor metastasis, expression of podoplanin and theVEGF-C/VEGFR-3signaling pathway by HE staining, immunocytochemistry, andso on. Results:1The basil group and group of mitomycin with high-dose basilcan extend the average survival days significantly,and the lifespan was greater than30%, the difference compared with salinenegative control group was significant (P<0.05), and the weightincreased significantly.2The high-dose basil group can inhibit in situ tumor of micewith H22liver cancer, the difference with the saline control groupwas statistically significant (P<0.05), but the differencecompared with high-dose and low-dose basil was not statisticallysignificant.3Immunohistochemical results showed that specific expression ofpodoplanin was in lymphatic endothelial cells, peritumoral lymphaticnumber of the high, medium and low dose of basil group, saline controlgroup were4.9±0.6,5.9±0.5,7.2±0.8and8.0±1.6,the high-dose and medium-dose of basil group compared with normal saline group,the difference was statistically significant (P<0.05).4. Staining results showed that protein expression of VEGF-C andVEGFR-3was located in the cytoplasm of tumor cells, were brownish-yellow granules, VEGFR-3can also be found in the vascular endothelialcells of cell stromal artery. The expression of tumor cells of high-doseand medium-dose basil were significantly lower than the saline controlgroup, the difference was statistically significant (P<0.05).Conclusion:1.Basil polysaccharide can make weight of mice with the model of H22lymphatic metastasis increased significantly, and prolonged the survivaltime.2Basil polysaccharide can inhibit in situ tumor of mouse with H22lymphatic metastasis, can reduce the number of peritumoral lymphaticvessels, inhibit the H22lymphatic metastasis by regulating VEGF-C/VEGFR- 3signaling pathway.
