Primary liver cancer is the malignant tumor of liver cells or intrahepatic bile duct epithelium. Hepatocellular carcinoma (HCC) is an aggressive cancer with a dismal outcome largely due to metastasis and postsurgical recurrence. Exploring the mechanism of the development of liver cancer and looking for new clinical therapeutic targets plays an important role in the treatment of hepatocellular carcinoma.Recent studies indicate that expression profiling with small,noncoding RNA gene products known as microRNAs(miRNAs) is a superior method for cancer subtype classification and prognostication. It is important to investigate the relationship between miRNAs and liver cancer invasion and metastasis.Purposes:Using miRNA microarray to detect the different expression of microRNAs in hepatocellular carcinoma/hepatocellular carcinoma with metastasis. By studying the correlation between miR-148a and metastasis, it can provide new ideas and experiment methods.Methods:MiRNA microarray was used to investigate the different expression of microRNAs in hepatocellular carcinoma/hepatocellular carcinoma with metastasis.We used transwell chamber assay and cell scratch test to detect the liver cancer cell line metastatic potential. The protein expression of matrix metalloproteinase2and9were evaluated by Western blot and gelatin zymography.Results:MiRNAs were differentially expressed in the miRNA microarray,one of the down-regulated miRNAs was miR-148a.Compared with the LV-miRNA group, LV-miR-148a group metastatic ability declined and expressed matrix metalloproteinase2and9lower than it.Conclusions:MiR-148a inhibits the invasion and metastasis ability of liver cancer cell by decreasing the expression of matrix metalloproteinase-2and metalloproteinase-9.
