Objective: To investigate the expression of MMP-3andepithelial-mesenchymal transition (EMT:Claudin-1、vimentin) in breastcancer,adjacent tissues and normal breast tissues.and analyze relationshipof MMP-3with epithelial-mesenchymal transition,and the role of MMP-3in tumor invasion and metastasis.Methods:Immunohistochemistry SABC was used to examine theexpressions of MMP-3, Claudin-1(epithelial marker) andVimentin(mesenchymal marker) in40cases of normal breast tissues,20cases of adjacent tissues,60cases of breast cancer tissues; then detectedthe relationship of mmp-3expression and metastatic axillary lymph nodes,and analyzed the connection of the three factors and the development、infiltration and metastas is of breast cancer.Results:Study found that:1)The positive rate of MMP-3in breastcancer was significantly higher than that in the adjacent tissue and normalbreast tissue (P<0.05).The positive rate of Claudin-1in cancerous tissue was lower than that in the adjacent tissue and normal breast tissue(P<0.05). The positive rate of Vimentin in breast cancer was significantlyhigher than that in the adjacent tissue and normal breast tissue(P<0.05);2)With the progress of the pathological stage of breast cancer,the expression of MMP-3increased gradually, simultaneouslyexpression of Claudin-1protein reduced and expression of Vimentinprotein increased.The spearman correlation analysis demonstrated thatMMP-3expression was negatively correlated with Claudin expression(r=-0.301,P=0.019),while it was positively correlated with Vimentinexpression(r=0.378,P=0.03).Conclusion:The results suggested that MMP-3may be mediatedepithelial-mesenchymal transition involved in the process of invasion andmetastasis of breast cancer.
