Objective: Periodontitis is a common disease. It is result in bleedinggums, periodontal overflow pus, loose tooth and even in tooth loss, and it isseriously influence the fuction of chew and digest. So far as it is the riskfactors of cardiovascular disease, diabetes, preterm birth. Periodontitis is amultifactor disease, plaque and bacteria is the mainly pathogenic factor.Sothe key to cure Periodontitis was to remove inflammation, and to promotethe regeneration in the destruction periodontal tissue, and to repair thenormal tooth function.In recent years, the scholars made a lot of researchabout Chinese herbal medicine treatment in Periodontitis,and found thatsome Chinese herbal medicine could kill the bacteria, and promote theperiodontal regeneration. We using the effective costituent of drynariaerhizoma, coptis chinensis and panax notoginseng(Burk)F.H.Chen to makethe chitosan thermosensitive hydrogel. We will study its effect onregeneration of periodontal defects and try to find a new way of theChinese herbal medicine to treatment Periodontitis.Methods:1To preparation chitosan thermosensitive hydrogel loadingsanhuangbu containing the appropriate proportion of drynaria fortunei naringin,berberine and panax notoginseng saponins.2To prepare a bony window on buccal aspect of the mandibular molarin36male SD rats. Then it were divided into three groups randomly,filledwith the chitosan thermosensitive hydrogel loading sanhuangbu were thetests, and filled with nothing or only filled with chitosan thermosensitivehydrogel were the controls.3In each group,animals were sacrificed after14or28days and thenthe mandibles were processed for histological examination andimmunohistochemical staining.The area of new bone and the opticaldensity(OD)of BMP-2was analysed and compared each groups.Results:1The area of new bone in test group were extremely larger than thatin the control group On14days and28days postoperation(P<0.05),andfilling with chitosan thermosensitive hydrogel were larger than in fillingwith nothing groups. Statistical difference was found among these groups.2After14,BMP-2protein expression level in the test group werestatistically higher expression than in the control group.Statisticaldifference was found in filling with nothing and filling only with chitosanthermosensitive hydrogel. After28,BMP-2expression level in the grouponly filling with chitosan thermosensitive hydrogel were statistically higherthan in the test group and in the blank control group.No statisticaldifference Was found in BMP-2expression between in the test group and in the blank control group.Conclusion:he chitosan thermosensitive hydrogel loading sanhuangbu caneffectively promote the periodonontal tissue regeneration.
