Objective: Study expression diversity of Pen-2, a component proteinof γ-secretase, at the CNS of various age groups of APP/PS1doubletransgenic mice, we try to determine the relations between Pen-2and AD.Methods: Genotyping the offspring of mated APP/PS1doubletransgenic mice, then using immunohistochemistry, doubleimmunofluorescence, Laser Co-focus and WB to observe the distributionand expression of Pen-2over cerebral cortex and hippocampus of1d,7d,21d,120d APP/PS1double transgenic mice and how Pen-2spread overentire CNS areas.Results: Pen-2is densely observed among1d,7d APP/PS1doubletransgenic mice and their wild-type cousins, through the growth of mice,Pen-2’s expression tends to decline; Pen-2immuno-positive reactions arestrengthened in the brains of APP/PS1double transgenic mice comparedwith those wild-types; Pen-2scatters widely around all CNS areas of adultAPP/PS1double transgenic mice, such as cerebral cortex, hippocampus, cerebellum, spinal cord, rhinencephalon, medulla and pons. Positive Pen-2cells at cerebral cortex and hippocampus outnumber positive Aβ cells andthey coexiste.Conclusions: Diversified distribution and expression of Pen-2appearin the brains of various age groups of APP/PS1double transgenic mice andtheir wild-types, and Pen-2is widely scattered around CNS of APP/PS1double transgenic mice, which suggests that yield of Aβ may be relatedwith AD.
