In this experiment, C57BL/6J high-fat diet mice were given purple sweet potato wine which was made in our laboratory. C57BL/6J mice were divided into7groups,every of which was10:the basic diet+water group (CL-W), the basic diet+alcohol group (CL-A), high-fat diet+water group (HF-W), high fat diet+alcohol group (HF-A), high-fat diet+anthocyanin solution group (HF-PC), high-fat diet+purple sweet potato wine diluent (is doubled dilution)(HF-WB) and high-fat diet+purple sweet potato wine off alcohol group (HF-DWB).In the end of the experiment, all mice were drawn blood from eyeball and were killed. Then we measured the content of total cholesterol (TC), triglyceride (TG), free fatty acid (NEFA), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C)and very low density lipoprotein cholesterol (VLDL-C) content in serum of mice and the activity of superoxide dismutase (SOD), malondialdehyde (MDA), total antioxidant capacity (T-AOC), catalase (CAT), glutathione peroxidase (GSH-Px) in serum and liver of mice, and we done biopsy of liver tissue and kidney tissue. All the results given a evaluation of purple sweet potato wine on the regulation effect of lipid metabolism, antioxidant activity, and protection of the liver, kidney of the mice on the high-fat diet.Results:the content of serum TC in HF-WB group was apparently reduced; the content of serum TG in HF-DWB group was significantly reduced; as for serum NEFA level, mice of HF-WB and HF-DWB groups were obviously reduced; the content of serum LDL-C in HF-WB group was evidently reduced; the VLDL-C level of serum in HF-DWB group was apparently reduced; however, serum HDL-C content showed no obvious difference.Antioxidant enzyme activity in serum and liver:SOD, T-AOC, GSH-Px vitality in HF-WB and HF-DWB groups were significantly improved; for the content of MDA, though the content of serum MDA in HF-WB and HF-DWB groups showed no evidently difference, the content of liver MDA in the two groups were obviously reduced; for the CAT activity in the serum and liver, HF-WB and HF-DWB group were not significantly difference, but they are much higher than the HF group by the data.Liver biopsy of the HF group showed severe diffuse fat steatosis; it of HF-WB and HF-DWB groups were more morphological normal, liver cell cord arranged in neat rows, cell plumped. Kidney biopsy of the HF group showed Pathomorphologically abnormal. Renal tubular epithelial cells began fusion. Tubular shriveled. Cytoplasm in different. Some of the cells broke apart. However, kidney biopsy of the HF-WB and HF-DWB groups were more morphological normal. Tubular was not shriveled. The lumen is clean. Renal tubular epithelial cells arranged in neat rows.Conclusion:(1) purple sweet potato wine have an inhibitory action on the weight gain of C57BL/6J mice on a high-fat diet.(2) the regulating effect of the wine on the abnormal lipid metabolism of the mice on high-fat diet is obvious.(3) antioxidant effect of the wine is also obvious, antioxidant defense system ability of the mice on high-fat diet is improved, the ability of cleaning free radicals is also improved, the regulating effect of the wine on the increasing of the vivo lipid peroxidation products is evident.(4) the liver and kidney damage caused by the high-fat diet would get better due to the wine.
