Objective To investigate whether the anti-platelet membrane surface GPIba monoclonal antibodies, SZ-2AN51are able to induce platelet apoptosis.Methods Platelet-rich plasma (PRP) and washed platelets were isolated from Venous blood of healthy volunteers, then washed platelets were incubated by SZ-2AN51antibodies at different times in37℃, the mitochondrial transmembrane potential (△ψm) depolarization level was detected by flow cytometry; And the expression of pro-apoptotic proteins bak and bad, anti-apoptotic protein bcl-xl and bcl-2, caspase-3protein was detected by Western blot.Results①SZ-2AN51antibodies time-dependent induce△ψm depolarization. When the platelets were incubated by the antibodies within1-3h, there’s no significant difference between the negative group(IgG、H1P1)and positive group(SZ-2、 AN51)(P>0.05); while since the4h incubation, which shows significant difference between the negative and positive group (P<0.05).②After6h incubation, the expression of pro-apoptotic proteins bak and bad, anti-apoptotic protein bcl-xl and bcl-2, caspase-3protein were detected by Western blot. SZ-2、AN51antibodies can increase the expression of bak and bad, decrease the expression of bcl-xl and bcl-2. caspase-3(32kd)③SZ-2, AN51antibodies are weak stimulation, which are not enough to cause PS exposure.Conclusion The anti-platelet membrane surface GPIba monoclonal antibodies, SZ-2. AN51can induce platelet apoptosis within a slight tendency.
