Objective: In this study, we aimed to detect the expression level of HIF-1α andGLUT-1in human normal and degenerated intervertebral disc tissues, theninvestigate the correlation between HIF-α and GLUT-1,the role of HIF-1αand GLUT-1may play in human lumbar intervertebral disc degenerationwas also studied.Methods: All patients diagnosed with intervertebral disc degeneration received MRIexamination, according to pfirrmann scale(I-V,fig1), they were dividedinto three groups:normal group/N(I,II),moderate degenerationgroup/M(III,IV) and serious degeneration group/S(V),the expression levelof HIF-1α and GLUT-1was detected by Immunohistochemistry andRT-PCR in their tissues. The experiments were taken from the nucleuspulposus tissue within the annulus, as much as possible to avoid theimmune effects of other substances on the spinal nucleus pulposus, whichcan more objectively reflect the real nucleus of immune status, theexperimental results the more reference value. P<0.05was considered toindicate a statistically significant difference,*P<0.05.Results: Immunohistochemistry showed that in normal human lumbar disc tissuesHIF-1α and GLUT-1showed a weak positive, moderate retreat expressionchanged group HIF-1a and GLUT-1is positive, severe degeneration group ofHIF-1α and GLUT-1expression was strongly positive. And with the degreeof lumbar disc tissue degeneration aggravated sexual expression alsoenhanced the three statistically significant differences (P <0.01). RT-PCRshowed that HIF-1α and GLUT-1gene is present in the human nucleuspulposus, and the disc degeneration and there is a correlation, statisticallysignificant differences between the three (P <0.01). Conclusions: HIF-1α and GLUT-1may participate in the process of humandegenerative lumbar disc tissue, there is a correlation between the expression;while HIF-1α and GLUT-1in human degenerative lumbar disc tissue has thesame increasing trend.
