Objective:Study the3,3’-Diindolylmethane on Eca109in vitro with radiotherapy sensitization effect and discuss the possible mechanism of action.Methods:1. Used CCK8method to measure different concentration DIM (10,20,40,60,80,100umol/1) on Eca109cell proliferation. Calculating the DIM on IC50Eca109, choose1/5IC50concentrations of DIM for subsequent sensitization experiments.2. Using clone formation experiment testing the1/5IC50concentrations of DIM of Eca109radiotherapy sensitization.3. FCM detecting group:the blank control group,6Gy radiation, dosing group (1/5IC50umol/L DIM) and drug+Radiation group (1/5IC50umol DIM/L+6Gy irradiation).Using FCM analysis above each cell cycle distribution.Results:1.CCK-8shows:different concentrations DIM (10,20,40,60,80,100umol/1) of Eca109after24h, OD value of each treatment group than the control group no significant decrease (P>0.05); the DIM effect on Eca109after48h,72h, OD value of each treatment group than the control group were significantly lower, statistically significant difference (P<0.05), the difference groups between each treatment group also statistically significant (P<0.05). According to DIM the inhibition rate of curve, we choose48h1/5IC50(12umol/L DIM) as experimental drug concentration of Eca109clone forming experiments.2. According to the result of clone formation experiment, Eca109cell inoculation efficiency is81.4%;The experimental group (1/5IC50, DIM+6Gy irradiation) radiotherapy sensitization ratio is1.33,3.FCM shows:other conditions without the intervention of Eca109cells in G0/G1period,second the S phase, the least of cell cycle ratio isG2/M phase. Give Eca109groups6Gy X-ray irradiation48h later (blank control group, pure dosing, pure light group, experimental group) compared with control group:The proportion of G0/G1phase increased significantly;S phase decreased significantly, the proportion of the G2/ M phase ratio increased significantly.The difference is statistically significant (P<0.05); Drug group than control group in G0/G1ratio increased significantly, have statistical significance,(P<0.05); Experimental group compared with drugs and radiation group the proportion of S phase decreased significantly, the Proportion of G2/M phase increased significantly, the difference has statistical significance,(P<0.05).Conclusion:DIM have radiotherapy sensitization effect to Eca109, the reason may be to block of esophageal Eca109cells in G0/G1phase of radiation sensitive to increase the radiation sensitivity, the next step to analyze the fine sensitization mechanism provides the experimental basis and theoretical basis.
