Objectives:To explore whether the gene abnormal expression of Juanskinases2(Jak2) or the abnormal phosphorylated of Jak2isassociated with systemic lupus erythematosus(SLE)complicated with thrombocytopenia.Methods:18patients of SLE complicated with immunethrombocytopenia who were admitted to YIJISHAN Hospitalfrom November2011to January2013were encolled into thestudy. Their peripheral blood(n=18) and bone marrow(n=15),clinical features and laboratory findings were collected.At thesame time18patients of primary SLE without thrombocytopeniawere selected randomly as primary SLE withoutthrombocytopenia control group. Their peripheral blood(n=18)and bone marrow (n=15) clinical features and laboratory findingswere collected.12patients of primary immunethrombocytopenic purpura were selected as primary ITP controlgroup. Their peripheral blood(n=12) and bone marrow(n=8), clinical features and laboratory findings were collected.12normalhealthy volunteers were recruited as healthy controls.Theirperipheral blood, clinical features and laboratory findings werecollected.The gene expression of JAK2mRNA in peripheral bloodand bone marrow was detectined with real-time polymerase chainreaction (RT-PCR).The realationship between the gene expressionof JAK2mRNA and the clinical features we analyzed.Thephosphorylated of Jak2was investigated by flow cytometry,and the realationship with the clinical features were also analyzed.Results:(1)The gene expression of JAK2mRNA in peripheral blood inprimary SLE without thrombocytopenia control group(0.652±0.401)was higher than that in primary ITP group(0.343±0.293),and this difference reached statisticalsignificance (P=0.009).The gene expression of JAK2mRNA inperipheral blood in SLE group was significant higher than that inSLE complicated with thrombocytopenia(0.319±0.313),andthis difference reached statistical significance (P=0.020).Andthe the gene expression of JAK2mRNA in peripheral blood inSLE group was related to peripheral platelet counts(P=0.030).(2)The gene expression of JAK2mRNA in bone marrow inprimary SLE without thrombocytopenia control group(0.683±0.265)was higher than that in primary ITP group(0.425±0.312),and this difference reached statistical significance(P=0.042).The gene expression of JAK2mRNAin bone marrowin primary SLE without thrombocytopenia control group(0.683±0.265) was significant higher than that in SLE complicated withimmune thrombocytopenia(0.288±0.264),and this difference reached statistical significance(P=0.000).The gene expression ofJAK2mRNA in bone marrow in SLE complicated with immunethrombocytopenia group was related to peripheral platelet counts(r=0.754,P=0.001)(3)The Mean fluoreseence intensity(MFI) of CD41a in primaryITP group (150.32±52.39)was higher than that in primary SLEwithout thrombocytopenia control group(121.56±59.35),and thisdifference reached statistical significance (P=0.039).(4)The Mean fluoreseence intensity(MFI) of Jak2in primarySLE without thrombocytopenia control group(103.28±24.59)washigher than that in SLE complicated with immunethrombocytopenia (79.56±36.77),and this difference reachedstatistical significance (P=0.047).Conclusions:1.The gene abnormal expression of Juas kinases2(Jak2)is associated with systemic lupus erythematosus(SLE)complicated with thrombocytopenia, and it has a correlationwith peripheral platelet counts.2. The machamism of Jak2may participates in pathopoiesis ofsystemic lupus erythematosus patients complicated withthrombocytopenia, and further study must be done to identifythe pathogenic mechanism.
