Objective:According to the proteomics research positive conclusion of the differential proteins by the former research group (the specificity proteomic study of endometriosis blood stasis syndrome type, National Natural Science Foundation, No.:30973770), immunohistochEMTical method was applied to test the expression of Annexin V, Vimentin, Myosin light chain3on eutopic and ectopic endometrium of observation group and on eutopic endometrium of control group;to further verify the EMTT specificity protein theory of blood stasis; to explore the molecule biological basis of blood stasis syndrome type; to discuss the relationship of the Annexin V, Vimentin and MYL3and the pathogenesis of EMTT; to be objectify and standardization of the traditional Chinese syndrome differentiation on the basis of experiment; to improve the accurate and curative effect of the traditional Chinese syndrome differentiation.Methods:The eutopic and ectopic endometrial tissues of EMTT patients were taken, in which6cases of blood stasis syndrome type,6cases of qi stagnation and blood stasis syndrome type,6cases of kidney deficiency and blood stasis syndrome type, and6cases of eutopic endometrial tissues of normal people. Using immunohistochEMTical method to study the expression differences of Annexin V, Vimentin and MYL3on eutopic and ectopic endometrium of EMTT patients and normal endometrial tissues, to test the above index expression on paraffin specimens in eutopic and ectopic endometrium of18EMTT patients and6cases of normal endometrial tissues, and to measure IOD values of the above result by Image pro Plus5.0software, and to analyse the expression differences on eutopic and ectopic endometrium and normal endometrial tissues among the EMTT blood stasis syndrome type, qi stagnation and blood stasis syndrome type, kidney deficiency syndrome type, blood stasis syndrome type patients by statistical methods.Results:(1) ImmunohistochEMTical studies suggest that the expression differences of Annexin V on eutopic and ectopic endometrium of blood stasis syndrome type and on ectopic endometrium of kidney deficiency and blood stasis syndrome type both have statistical significance (P<0.05); the expression differences of Annexin V on eutopic and ectopic endometrium of three syndrome types compared with normal control group have statistical significance (P<0.05);(2) the expression differences of Vimentin on ectopic endometrium of blood stasis syndrome type compared with ectopic endometrium of kidney deficiency and eutopic endometrium of qi stagnation and blood stasis syndrome type and normal control group all have statistical significance (P<0.05); the expression differences of Vimentin on ectopic endometrium of three syndrome types compared with normal control group have statistical significance (P<0.05);(3) the expression differences of MYL3on ectopic endometrium of three syndrome types compared with normal control group have statistical significance (P<0.05); the expression differences of MYL3on ectopic endometrium of three syndrome types have no statistical significance (P>0.05);Conclusion:(1) the significant expression differences of Annexin V, Vimentin and MYL3three protein on eutopic and ectopic endometrium compared with normal control group prompted that there may be a certain relationship of Annexin V, Vimentin and MYL3with EMTT, and further verified Annexin V, Vimentin and MYL3are the three indicators for EMTT specificity protein markers;(2) Annexin may be the essence of blood stasis syndrome type for which has expression differences EMTT patients of blood stasis syndrome type compared with normal control group, through the blood stasis syndrome type and the other syndrome types had no significant differences expression. The significant expression differences of Vimentin on ectopic endometrium of blood stasis syndrome type except qi stagnation and blood stasis syndrome type, thus it may be related to the blood stasis syndrome type even its pathogenesis;(3) MYL3has no significant differences expression in blood stasis syndrome type and other syndrome types syndrome type, so MYL3can not be concerned as EMTT specific protein of blood stasis.
