Experimental Study On The Pharma-odynamics Of CXDT Grains In Treatment Of Migraine

Objective:This study aims to know the effect and possible mechanism of Chuangxiong Dingtong Grains (CXDT Grains) in treating migraine.Methods:1. The anti-migraine effect of CXDT was studied with migraine rat model caused by subcutaneous injection NTG and migraine mice model caused by reserpine.2. The effect of promoting blood circulation of CXDT Grains was studied with auricle microcirculation mice and blood stasis rat model.3. The annagesis effect of CXDT Grains was studied with hot plate inducing algesia and acetic acid inducing writhing response.4. The effect of sedative of CXDT Grains was studied with locomotor activity frequency of mice in5minutes, sodium pentobarbital sleeping time in mice and sodium pentobarbital mice sleep number.Results:1. The CXDT Grains can significantly improve the active substance in rat plasma and brain neurotransmitter content of migraine rat model caused by subcutaneous injection NTG. It can significantly improve the content of5-HT, NE, ET in plasma and that of5-HT, DA in the brain tissue. It is possible to reduce the content of NE in the brain tissue. To the reserpine mice, CXDT Grains can also significantly improve the content of5-HT in serum and brain tissue, reduce the content of DA, NE in brain tissue.2. CXDT Grains can significantly accelerate the microvascular flow rate of auricle microcirculation, expand its microvascular vein caliber and micro-artery diameter, increase the amount of open capillaries, significantly reduce the whole blood viscosity and the high shear blood relative index of blood stasis rat mode. On the other hand, CXDT Grains can reduce the trend of the whole blood relative index.3. CXDT Grains can’t increase the pain threshold in mice in the hot plate test, but it can significantly reduce the writhing caused by intraperitoneal injection the glacial acetic acid.4. CXDT Grains can significantly reduce the locomotor activity frequency of mice in5minutes, extend the threshold dose of sodium pentobarbital sleeping time in mice, and increase the threshold dose of sodium pentobarbital mice sleep number. Conclusions:CXDT Grains have the effect of activating blood circulation, analgesia, calming the liver and subduing yang, anti-migraine. These effects can be achieved mainly by adjusting the level of the brain neurotransmitters and vasoactive substances, improving microcirculation and blood.
