ObjectiveTo explore the impact of estrogen on adipogenic differentiation of long-termcryopreservation and the fresh kidney fat capsule adipose-derived mesenchymalstem cells(KFC-ADSCs).MethodsThe third generation of long-term cryopreserved and the fresh KFC-ADSCswere divided into four groups, and all of which were induced to adipogenic cellsby induced fluid,which content dexamethasone, insulin, indomethacin,isobutylmethylxanthine. Among them,the long-term cryopreserved KFC-ADSCs wasdivided into group A[given low-dose estrogen (10-7M))] and group B(only giventhe induced fluid).The fresh KFC-ADSCs were divided into group C and group D,group C was given low-dose estrogen (10-7M) and induced fluid to induceadipogenic differentiation, group D was only given to induce adipogenicdifferentiation the induced fluid.ResultsThe morphology of the third generation of long-term cryopreserved and freshKFC-ADSCs both were fusiform, polygonal, showing whirlpool arragement, therewas no significant difference between the third generation of long-termcryopreserved and fresh KFC-ADSCs in morphology and cells arragement; Cell Surface antigen CD29, CD44were all detected positiveexpression, but CD31wasall detected negative expression; Intracellular lipid droplets in the cells wereobserved after adipogenic differentiation by oil red O staining, the cells all showedpositive oil red O staining; The Adipogenic volume comparison of each groupafter adipogenic differentiation was detected in the fourteenth day [TheAbsorbance value(value A)which was measured by spectrophotometer wasselected as comparative target].Compared with the group A, the Absorbancevalue of the group B was significant higher(P<0.05);compared with the group C,the Absorbance value of the group D was significant higher(P<0.05). But nosignificant difference was detected in group A and group C (P>0.05).ConclusionsLow-dose estrogen can inhibit the adipogenic differentiation of long-termcryopreservation KFC-ADSCs. There was no significant difference between thethird generations of long-term cryopreserved and the fresh KFC-ADSCs onadipogenic differentiation.
