ObjectiveInvestigate the influence and protection actions of oxidative stress afteradministration different phosphodiesterase inhibitors such as Milrinone. indiabetic rats. Discuss the protection effect on the complications of heart, brainand kidney in diabetes rats. Provide reference for drug prevention and treatmentdiabetic complications such as cardiovascular diseases.Methods30rats were randomly divided into normal control group, diabetes controlgroup, Milrinone group, theophylline group and sildenafil group,6in each group.Rats became diabetic by a single administration of streptozotocin. After15-dayssuccessive treatment with milrinone, sildenafil and theophylline respectively tothe diabetes rats, the blood glucose level and weight was tested in this period.Then the animals were sacrificed. The levels of malonedialdehyde(MDA), Totalantioxidant capacity (TAOC), Superoxide Dismutase(SOD) and4-hydroxynoneal(4-HNE) were estimated in plasma or cardiac muscle, renal tissue andbrain in rats. The characteristic of4-HNE expressed in renal and brain tissuewere checked with the immunohistochemisty method. The quality of4-HNE inrenal and cardiac tissue were tested with the western-blot method.ResultsCompared with the normal rats, a significant increase in the levels ofplasma glucose, MDA and4-HNE were found in the DM group. Concomitantdecrease in the levels of TAOC, SOD was observed in diabetic rats’ plasma(P﹤ 0.01). These alterations MDA,4-HNE, TAC and SOD in plasma/cardiac muscle,kidney, brain tissue were reverted back more or less to the normal level after thetreatment with milrinone(P>0.05). But the glucose level was no obvious changeafter treatment of milrinone(P﹤0.01). The immunohistochemisty results showed4-HNE expression was obvious the rats’ brain and kidney in diabetes group butit was weaken in the milrinone group. The diabetic rats by PDEIs reduced4-HNE expression by western-blot method in cardiac and renal tissue(P﹤0.05).ConclusionsDiabetes rat model could be induced by streptozotocin. PDEIs milrinonecould decrease the MDA,4-HNE level and increase TAOC, SOD level.Antioxidant activities of milrinone may contribut to the protection againstoxidative stress in streptozotocin-induced diabetes.
