Estrogen is a kind of steroid that play essential roles in the female reproductive system,stimulate secondary sexual characteristics, and regulate bone growth and maturation. It isproduced by the ovaries and placenta. A drenal can also produce a small number of estrogens.Dysregulation in estrogen metabolism has been implicated in infertility, obesity, and severalhuman diseases, including breast cancer, prostate cancer, Alzheimer’s disease. Estrogens areused as pharmaceuticals to cure the gynecological diseases. However, estrogens can furtherthe growth of animals and have an impotance impact on the metabolism of protein, sugar andgrease. So estrogens is used as an additive in fodder, cosmetic and food. Estrogens threats tohuman health through the accumulation of the food chain. It may cause the precocious pubery,teratogenic and carcinogentic. Therefore, it has great significance to determinate estrogens.Chemiluminescence(CL) analysis owns the advantages that sensitivity, simplity andfacility. It is widely used in pharmaceutical analysis, environment monitoring and foodanalysis ect. Electrochemiluminescence(ECL) is developed from the electrochemical analysisand chemiluminescence. As one branches of chemiluminescence, electrochemiluminescenceanalysis is studied in recent years. It has some good points that surpass thechemiluminescence, such as the controllable of the reaction and location, little backgroundinterference. The research sections were as following:Chapter1: IntroductionFirst, the principle and common systems of chemiluminescence and ECL method andapplications of them in analysis have been summarized. Furthermore, we have described thequantum dot(QD) and its application. At last, the purpose and significance of this dissertationwere presented.Chapter2: The study of chemiluminescence system and its applicationUsing KMnO4-Ru(phen)3Br2-Na2SO3chemiluminescence system to determine estradiol.It is found that in acid conditions, KMnO4can oxidate Ru(phen)3Br2and produce weakchemiluminescence. Estradiol in the presence of sodium sulfite can strongly enhance thesystem’s signal. The linear ranges of6.0×10-108.0×10-8g·mL-1, the detection limit areestimated to be3.8×10-10g·mL-1. It is used successfully in the determination of the actual sample.Then, in alkaline solution, luminol-K3Fe(CN)6system can produce weakchemiluminescence. CoDP(metalloporphyrin) could enhance the CL signal. Acetaminophencould inhibit the signal. Based on it, the new CL methods were developed to thedetermination of acetaminophen. The linear range of4.0×10-102.0×10-8g·mL-1, the detectionlimit is3.0×10-10g·mL-1. Paracetamol Tablets is determinated.Chapter3The study of ECL and its appilicationIt is found that luminol-H2O2ECL system has an weak signal at+1.7V. CdS and ZnSequantum dot can enhanced the signal. Bisphenol A and diethylstilbestrol can inhibit the ECLsignal. Based on this, the new ECL methods were developed to determinate Bisphenol A anddiethylstilbestrol. The linear range were of2.010-84.010-7mol L-1,1.010-81.010-7g·mL-1, the detection limit were1.510-8mol L-1,8.010-9g·mL-1respectively. At last,associate with flow injection analysis, we developed a ECL system that CdS QD-Luminol.Catechol can inhibit the signal. Under the optional conditions, the linear range of8.010-81.0×10-6g mL-1, the detetion limit was5.5×10-8g mL-1.Chapter4: Conclusion and prospect.
