Trichosanthes kirilowii Maxim is a kind of herbaceos perennial of Cucurbitaceae.Trichosanthes Linn. and Trichosanthis pericarpium is the desiccative pericarp of ripe fruit ofTrichosanthes kirilowii Maxim. or Trichosanthes rosthorinii Harms. Recent studies show thatTrichosanthes pericarpium has various biological activities such as antibiotic, antituberculotic,expectorant, antidiabetic, antineoplastic and anti-inflammatory activities. Trichosanthespericarpium is of rich resoures, low prices, little side effects and high developing and utilizingvalue.Recent years, though the researches of pharmacological effects of Trichosanthespericarpium is increasing, the effective constituents from Trichosanthis pericarpium whichmatches the functions of clinical use is not clear. So for further research of pharmacologicaleffects and the development of Trichosanthis pericarpium, we first studied the chemicalconstituents of Trichosanthis pericarpium, which can be applied to the qulatity evaluation ofTrichosanthis pericarpium.On the based objective of the present study, the relevant literatures of Trichosanthes Linn.,including the literatures of constituents, pharmacological effects and quality evaluation werefirst collected. Isolation and purification of chemical ingredients from Trichosanthispericarpium were conducted by silica gel column chromatography, sephadex columnchromatography, C18reversed-phase chromatography, high-speed counter-currentchromatography and prepared high performance liquid chromatography.18compounds weresuccessfully isolated from Trichosanthis pericarpium. The structures of the compounds wereidentified by ESI-MS and NMR. We also established the method using high performanceliquid chromatography with hydrophilic column to determine the content of adenosine fromTrichosanthis pericarpium. Ultrasonic extraction was used to extract adenosine fromTrichosanthis pericarpium. Adenosine was separated on Waters XBridge Amide column witha gradient elution using acetonitrile-20mM ammonium acetate-0.2%ethylic acid solution asmobile phase. Linear regression of adenosine, accuracy of chromatographic system, stabilityof Trichosanthis pericarpium samples, repeatability of the method and average recovery werestudied.18compounds were successfully isolated from Trichosanthis pericarpium, and amongwhich13compounds were successfully identified. The13compounds were elucidated aspalmitic acid (1), α-spinasterol (2), β-Sitosterol (3), stigmasterol (4), 4-methoxy-3-hydroxy-benzoic acid (6),quercetin-3-O-α-D-nucleoside (8), vanillicacid-4-O-β-D-glucopyranoside(10), quercetin-3-O-β-D-glucopyranoside (11),chrysoserid-7-O-β-D-glucoside (12), luteolin7-O-β-D-gulcoside (13), adenosine (14), vanillicacid (15), quercetin-3-O-β-D-rutinoside (16), among which compound6,8,10were fistisolated from Trichosanthes Linn., compound11,13and16were first isolated fromTrichosanthes kirilowii Marim..The study established the method using high performance liquid chromatography withhydrophilic column to determine the content of adenosine from Trichosanthis pericarpium.Results showed that the method had good precision, recovery and can be used fordetermination of adenosine from Trichosanthis pericarpium. This work provided a newmethod for the rapid quality control of Trichosanthis pericarpium.The research work and the results mentioned above have great significance to furtherstudy of pharmacological effects, the development of Trichosanthis pericarpium and qualityevaluation of Trichosanthes kirilowii Marim.. |
