Objective:To investigate the changes of BMP-7and TGF-J31expression in brain tissue following cerebral ischemic reperfusion in SD rat, explore the neuro protective efficacy and mechanisms of the Danhong injection on brain protection.Methods:1. Animal Groups and models:Sixty healthy male SD rat (250g-320g,10-12weeks old, CLA) were randomly divided into four major groups:Normal Control Group (n=10), Sham-operated Group (n=10), Ischemia-Reperfusion Group6hour (I/R6h Group, n=5) Ischemia-Reperfusion Group1day (I/Rld Group, n=5), Ischemia-Reperfusion Group3day (I/R3d Group, n=5), Ischemia-Reperfusion Group7day (I/R7d Group, n=5)and Danhong injection Group6hour (I6h, n=5), Danhong injection Group1day (Ild, n=5), Danhong injection Group3day (I3d, n=5), Danhong injection Group7day (I7d, n=5). The middle cerebral artery ischemic reperfusion mode was made by using the Longa’s method. Based on the Berderson’s, we evaluate the nerve function after cerebral ischemic reperfusion. The animals were sacrifice by decapitation at correspondence time.2. Hematoxylin and eosin stain was used to observe the morphological changes of hippocampus o3.1mmunohistochemical method was applied to detect the expression of BMP-7and TGF-β1level.4. Data were analyzed with SPSS11.0. Results:1.The successful rate of the models was80.6%.2.HE stain:The rats had normal structure and morphology of neuron in brain tissues in Sham-operated Group and Normal Control Group. Degeneration and necrosis of neuron occurred, which exacerbated at6hour,1day,3day,7day, with hyperplasia of neuroglial cells in I/R Group. In corresponding sub-groups, the brain damages in Treatment Group were slighter than I/R Group.3.Immunohistochemical results:In Normal Control Group and Sham-operated Group, there was low BMP-7and TGF-β1expression, mainly in neurons and neuroglial cells. Both gradually raised after ischemic reperfusion, in which BMP-7reached a peak concentration1day later and then declined at3day and7day while TGF-βshowed decline following a Day3peak. The performances in Treatment Group were similar to I/R Group, however, with increase to different degrees at all time points.Conclusions:Danhong Injection could exert neuroprotective effects by promoting the expression of BMP-7and TGF-β1.
