Objective:To observe the morphological characteristics of tissue pathology of suspected parasitic samples. To detect samples infected with sparganosis.Methods:19specimens of suspected parasitic samples and clinical materials from Changsha, Huaihua, Shaoyang, Xiangtang were collected. pathological features was obtained by light microscope. Immunohistochemistry was used in detecting the antigen of sparganosis; Genomic DNA was extracted from the two suspected cases and two no-worm samples which were positive for the antibody. PCR was used to amplify target sequence. Target band was observed by electrophoresis and sequencing. Obtain the coincidence rate between sequence of two no-worm cases and known gene through the Blast. PCR methods with universal pairs were used in detecting the other worms.Results:Six cases were diagnosed as sparganosis definitely and two were suspected as sparganosis; Other worms emerged in the seven cases, one of the which was caused by heterophyid. There were no worm in the four cases, two of which had serological results. positive reaction was found on two suspected cases and two no-worm cases with no serological results. The target band of Spirometra erinaceieuropaei from the two cases showing positive reaction to the serum also was obtained by PCR amplification from the two suspected cases and the two positive cases with no worm. The targed bands of the two positive cases with no worm amplified by F650/R800was100%matched to the known sequences in the pubmed. The band of the one case amplified by F800/R1120was97%matched to the known sequences in the pubmed and the other was100%. There were negative results in detecting the samples of other worms by PCR using universal pairs.Conclusion:The study describe the histopathological feature of complicate samples of sparganosis and explore the dilemma of diagnosis of parasitic pathology. The serum of Spirometra erinaceieuropaei immuned rabbit possesses a value of application in detecting suspected samples. PCR test can play an important role in detecting no-worm saparganosis samples.17figures,1tables and32references...
