Objective:To explore the influence of uterine-shaped silica gel device on the endometrium of goats by observing the local pathological changes in endometrium tissue following the insertion of the device. Methods:Materials:uterine-shaped silica gel device (patent device) and metal uterine-shaped ring. Nine goats of reproductive age were randomly divided into3groups:silica gel group (two of the patent devices were inserted into each of the goat uterine cavity after the hysteroscopic biopsy of the endometrium), uterine shaped ring group (two of the metal ring devices were inserted into each of the goat uterine cavity after the hysteroscopic biopsy of the endometrium), and control group (no devices were inserted into the cavity after the hysteroscopic biopsy of the endometrium). On day7,30and90following the insertion, hysteroscopic endometrium biopsy were carried out, and the endometrial tissues were observed by light and electron microscope for the morphologic variations. At the same time, the expression of inflammation cytokines IL-1β and TNF-a were examined by immunohistochemistry. Results:On day7and30following the insertion, neutrophil infiltration, mitochondrial vacuolization, and cytoplasmic edema were visible in both experimental groups. The expression of IL-1β and TNF-a were increased in these groups and had significant statistical difference compared to the control group (P<0.05). However, on day90following the insertion, very few neutrophils were seen on the light microscope. Mitochondrial vacuolation were not clearly visible on electron microscope, only slightly cytoplasmic edema was seen. Neutrophil infiltration and mitochondrial vacuolization between three groups failed to show significant difference (P>0.05). Expressions of IL-1beta and TNF-alpha were low in all groups and the levels had no significant difference (P>0.05). Conclusion:Compare to the uterine-shaped rings, which are commonly used in clinical treatment of intrauterine adhesions, patent uterine-shaped silica gel IUD is preliminarily confirmed biological safe.
