Objective:We expect to construct a novel membrane in order to minimize or avoid the use of anticoagulation and be suitable for the high-risk of bleeding patients. In order to construct a new type of hemodialysis membrane which can provide extraordinary biocompatibility and more efficient function of anticoagulant, we plan to graft argatroban to the polyether sulfone membrane by blending technology and the theory of phase transformation.Methods:(1) Preparation of argatroban modified polyether sulfone membrane.①Preparation of argatroban modified polyether sulfoneaccording to the principle of polymer blending modification, using the polyether sulfone material as base material, adopt the method of nitration and nitro reduction, preparing amino modified polyether sulfone material, and then under the action of EDC catalyst, grafte argatroban to amino polyether sulfone materials, we weighed the polymer’s weight before and after grafted, which are denoted as WO and W1respectively. Then the graft-ratio (G%) is calculated according to the formula which is G%=(W1-W0)/W0*100%; Characterization of prepared functioned polymers by FTIR and1HNMR spectra through the whole process of chemical reaction each steps.②Preparation of argatroban modified polyether sulfone flat-sheet membrane Blending the polyether sulfone and argatroban modified polyether sulfone, then adopt the method of phase transformation to prepare the modified polyether sulfone membrane; To characterize the surface of the prepared membrane, XPS was employed to examine the surface compositions of the prepared PES and PES-Arg membrane. And SEM was used for observation of membrane surface morphology. (2) The activated partial thromboplastin time (APTT), prothrombin time (PT) and thrombin time (TT) were conducted to detect the anticoagulation performance of the modified membrane.Results:(1) Preparation of argatroban modified polyether sulfone membrane①Preparation of argatroban modified polyether sulfoneAfter chemical reaction, the grafted-ratio respectively was53.74%,44.73%and59.18%, which proved that there are something on the membrane.a. FTIR spectra of PES-NO2, the peak at1533.92cm-1was attributed to the asymmetric stretching vibration of-NO2, the peak at1325.16cm-1was attributed to the symmetric stretching vibration of-NO2, and the peaks at901.86cm-1and855.25cm-1were assigned to the C-N stretching. This show-NO2has successfully grafted on the polyether sulfone.1HNMR spectra for the PES-NO2, a peak at δ=8.840(s,2H) was observed, which was attributed to the-NO2grafted onto the PES main chain, due to the electron withdrawing effect of the grafted-NO2.b. FTIR spectra of PES-NH2, the peaks at3362.07cm"1,3442.70cm-1and1622.44cm-1were attributed to the N-H stretching, and the peak at1285.35cm-1was attributed to the C-N stretching. This show-NH2has successfully grafted on the polyether sulfone.1HNMR spectra for PES-NH2, the peaks at δ=7.25(d,2H),δ=7.00(d,3H) and δ=6.78(d,3H) demonstrated the successful nitro-reduction reaction.c. FTIR spectra of PES-Arg, the peaks at3441.39cm-1were the chemical shifts of the phenyl on the PES main chain, the peaks at1433.13cm-1were attributed to the C-S-C stretching, the peaks at1627.48cm-1were attributed to the-CO-NH-stretching, which was the Characteristics of the argatroban and PES-NH2reaction products,it shows that argatroban has been successfully grafted to the polyether sulfone;1HNMR spectra for PES-Arg, the peaks at δ=7.27(d,2H),δ=7.04(d,3H) and δ=6.98(d,3H) demonstrated the successful nitro-reduction reaction.Furthermore, the peaks at δ=6.60(d,2H),δ=5.29(s,1H) and so on suggests that argatroban has been successfully grafted to the polyether sulfone.②Preparation of argatroban modified polyether sulfone membrane The modification process was successfully confirmed by detecting XPS showed that one kinds of particular newly chemical elements N was detected on the surface of the modified membrane. The results of SEM showed that morphology characteristics and appearance of modified surface had significant change.(2) anticoagulant properties testPT, APTT and TT test of modified membrane proved that argtroban modified membrane have better anticoagulation characteristics than polyether sulfone membrane.Conclusion:(1) the blending technology and the principle of phase transformation can be successfully modified polyether sulfone membrane.(2) argatroban modified polyether sulfone membrane has good anticoagulant properties...
