Objective:Ivestigate the expression of vascular endothelial growth factor (VEGF) and soluble fms-like tyrosine kinase receptor1(sFlt-1) in serum and villus of the pregnant women and its association with the unexplained early spontaneous abortion (UESA)Methods:40women who had missed abortion and40women who asked for induced abortion were selected respectively to be the experimental group and the normal group.(1) The enzyme linked immunosorbent assay (ELISA) was used to evaluate the pre-operation serum levels of VEGF and sFlt-1.(2) Real-time polymerase chain reaction (RT-PCR) was used to evaluate the expression of VEGF mRNA and sFlt-1mRNA in villus.(3) Compare the serum levels of VEGF and sFlt-1&the expression of VEGF mRNA and sFlt-1mRNA in villus, then we analysed the correlation between the experimental results.Results:1. The serum level of VEGF in the expreimental group (127.64±15.28pg/ml) was significantly lower than that in the normal group (160.05±18.02pg/ml), P<0.05; The serum level of sFlt-1in the expreimental group (6522.01±940.62pg/ml) was significantly higher than that in the normal group (4765.05±721.80pg/ml), P<0.05.2. The expression of the VEGF mRNA in the villus of the expreimental group (0.0227±0.0047) was significantly lower than that in the normal group (0.0934±0.0150), P<0.05; The expression of the sFlt-1mRNA in the villus of the expreimental group (0.0662±0.0103) was significantly higher than that in the normal group (0.0230±0.0042), P<0.05.3. The serum levels of VEGF were correlated with the expressions of VEGF mRNA in the villus both in the expreimental group(r=0.798,P<0.05) and the normal group (r=0.710, P<0.05); The serum levels of sFlt-1were correlated with the expressions of sFlt-1mRNA in the villus both in the expreimental group (r=0.584, P<0.05) and the normal group (r=0.649, P <0.05); The serum levels of VEGF were correlated with that of sFlt-1both in the expreimental group (r=-0.688, P<0.05) and the normal group (r=-0.762, P<0.05); The expressions of VEGF mRNA in villus were correlated with that of sFlt-1both in the expreimental group (r=-0.614,P <0.05) and the normal group (r=-0.655,P<0.05)Conclusions:1. The reduced expression of VEGF may be correlated with the early spontaneous abortion.2.The abnormal elevation of expression of sFlt-1may antagonize the effect of VEGF and participate in the early spontaneous abortion.
