CK18OD value in serum samples of colorectal cancer patients pre-and posteoperatively were checked out by enzyme-linked immunosorbent assay, and the EliVisionTM Plus immunohistochemical technique was used to detect N-cadherin, CDX2, P120ctn in colorectal cancer. It aims to study below aspects:To explore the changes and the clinical significance of CK18in colorectal cancer patients preoperatively and posteoperatively in the serum. To investigate the expression of N-cadherin, CDX2and P120ctn in human colorectal carcinoma, and to evaluate the clinical significance of these two markers in the histologic classification, invasion, metastasis and prognosis of colorectal carcinoma. To search for the relations among N-cadherin, CDX2, P120ctn in colorectal cancer and CK18in blood.Sixty cases of colorectal cancer and20cases of paracancerous normal tissues were collected from the department of Pathology in the251st hospital of PLA ranging from May in2010to March in2011. The expression of N-cadherin, CDX2, P120ctn were detected in colorectal carcinoma using immunohistochemistry technique of EliVisionTM Plus. CK18OD value in serum samples of colorectal cancer patients pre-and posteoperatively were checked out by enzyme-linked immunosorbent assay. And the relationships among the four markers and the age, gender, the size of tumor, the degree of differentiation, TNM stage, the depth of invasion, metastasis of lymph nodes was analyzed, also the relationships among the four markers was analyzed.The CK18OD value of the whole group of60patients had a certain degree of reduction, in which24patients exhibited a significant decreased by more than50%, the differentce of CK18OD values in the different TNM stages groups of patients preoperatively and posteoperatively had significant difference(P<0.05). In patients serum, the CK18contents in UICC stages I and IV was higher than stages II and III. There was not significant difference in patients serum between high differentiation adenocarcinoma and poor differentiation adenocarcinoma (P>0.05). The positive rate of N-cadherin in normal tissue was10%(2/20), the positive rate of N-cadherin in colorectal cancer was65%(39/60). The positive rate of N-cadherin in colorectal cancer was significantly higher than that in normal tissue(P<0.05), The positive rates of N-cadherin were25%,40%,87.5%in well, moderately, poorly differentiated colonic adenocarcinoma; the expression of N-cadherin among groups had statistical difference(χ2=9.514, P=0.002). The positive rate of N-cadherin correlate with the depth of invasion, lymph node metastasis, the TNM stage, while not correlated with age, gender and the size of tumor.The positive rate of CDX2in normal tissue was95%(19/20), the positive rate of CDX2in colorectal cancer was65%(39/60), the positive rate of CDX2in colorectal cancer was significantly higher than that in normal tissue(P<0.05). The positive rates of CDX2were83.3%,72.5%,25%in well, moderately, poorly differentiated colonic adenocarcinoma, the expression of N-cadherin among groups had statistical difference (χ2=6.388, P=0.011). The positive rate of CDX2correlate with the depth of invasion, lymph node metastasis, the TNM stage, while not correlated with age,gender and the size of tumor. The positive rate of P120ctn in normal tissue was100%(20/20), the positive rate of P120ctn in colorectal cancer was66.7%(40/60), the positive rate of P120ctn in colorectal cancer was significantly higher than that in normal tissue(P<0.05). The positive rates of P120ctn were91.7%,67.5%,25%in well, moderately, poorly differentiated colonic adenocarcinoma, the expression of P120ctn among groups had statistical difference(χ2=8.995, P=0.003). The positive rate of P120ctn correlate with the depth of invasion, lymph node metastasis, the TNM stage, while not correlated with age, gender and the size of tumor. The negative correlation between N-cadherin and CDX2(r=-0.500, P<0.01), and the negative correlation between N-cadherin and P120ctn (r=-0.519, P<0.01), and The positive correlation between CDX2and P120ctn (r=0.735, P<0.01).CK18might be used as a biomarker determing the prognosis of colorectal cancer patients postoperatively and provided a reference for clinical chemotherapy. N-cadherin, CDX2and P120ctn may play a cooperative role in the onset of malignant progression of colorectal cancers, and correlated with epithelial-mesenchymal transition significantly, which were important biological markers in the infiltration and early metastasis of colorectal cancer. Co-detection of N-cadherin, CDX2and P120ctn might be important to evaluate the metastasis and prognosis of colorectal carcinoma.
