Aim:Fufang Danshenfang is one of most used TCM formulae in eastern counties. However, the quality of this formula is difficult to control according to its complex composition. In the present study, we aim to develop This paper intends to develop a composition of effective components extracted from Fufang danshenfang which may act much more effective than the Fufang danshenfang on myocardial injuries.Method:A model of MIRI was established in rats induced by ligating of the left anterior descending of the coronary artery (LAD). Through the determination of serum super oxide dismutase (SOD), lactate dehydrogenase (LDH) and aspertate aminotransferase (AST), myocardial infarction area by NBT staining method, HE dyeing histopathological examination, hemodynamic parameters, detection of cell apoptosis by TUNEL staining method and the study of apoptosis related gene Bcl-2, Bax, the effect of FDT and XMLT on MIRI was investigated, and its mechanism was also discussed. The combination of active ingredients was selected on the model of acute myocardial ischemia induced by pituitrin.Results:Combination of ginsenoside Rgl/tanshinone II A/dios-genin can reduce the level of LDH, AST, MDA myocardial ischemia rats induced by pituitrin (PIT), increase the activity of SOD in MIRI rats; FDT and XMLT reduced the release of myocardium LDII and AST, increased the activity of SOD in serum significantly; Infarct size was decreased; The histological changes of hearts were less severe; Apoptosis index was reduced, decreased the Bcl-2/Bax ratio. Improved heart function through increasing SBP, DBP, MAP, LVSP, and±dp/dtmax in MIRI rats. In summary, the protective effect of FDT and XMLT are involved in itsConclusion:Collectively, these results manifested that the actions of scavenging oxygen radical and inhibitory effect on ceil apoptosis may contribute to explain the heart-protective effect of FDT and XMLT. Combination of ginsenoside Rgl/tanshinone Ⅱ A/diosgenin can resistant acute myocardial ischemia. Furthermore, this research provides new information to the study of these drugs, which is helpful to reveal the heart-protective material base of Compound Danshen Formula. At the same time, it has made the beneficial attempt to the development of the new compound of Chinese traditional medicine (TCM). And it’s feasible to explain the theory of prescription compatibility by the method of modern pharmacology.
