baicalein as a naturally occurring flavonoid compounds of Scutellaria baicalensis Georgi,It is widely biological activities and pharmacological actions because of its anti-bacterial,Anti-tumor, Anti-virus, protect liver and other wide availability characters. In recent years,there are many studies have investigated the effect of the antibacterial activities, but theantibacterial mechanisms were reported less. Baicalein (BAI) is an effective bactericide.Inthis study, the antibacterial activity and mechanism experiments of baicalein on MRSA werecarried out by determining conductivity and content of macromolecules of membranepenetrability, protein synthesis changes and the inhibition of DNA topoisomerase activitiesand manner of the effect of baicalein on DNA. The results are as follows:(1) This paper studies that antibacterial activity of baicalein on MRSA by utilizingOxford plate and TTC Test. The effect of baicalein to against MRSA was obvious and theminimum inhibitory concentration of baicalein is0.04mg/mL.(2) By scanning electron microscopy, determination of electrical conductivity andbiological macromolecules changes (DNA and RNA) to study the effect of baicalein on thepermeability of cell membrane. The experiment showed that baicalein did not damage theintegrity of the cell membrane, which was only a minor damage to the cell membrane.(3) The results of SDS-PAGE of experiment research showed that the expression ofproteins were significantly decreased44.62%than that compared to control group withbaicalein for16h, which suggested that baicalein inhibited the synthesis of bacterial proteins.(4) The agarose gel electrophoresis results showed that baicalein can inhibit the activityof DNA topoisomeraseⅠ andⅡ indose-dependent manner. The activity of topoisomeraseⅠandⅡ was inhibited at0.05mg/mL of baicalein, with complete inhibition of topoisomerase Iand II activity at0.2mg/ml.(5) To investigate the effect of baicalein on DNA by using the UV-absorptionspectrometry and the agarose gel electrophoresis. The results of this experiment appearedhypochromic effect and red shift, and the stronger was the intercalation, the more obvious wasthe hypochromic effect. The results indicate that the baicalein can intercalation of DNA, andhave a direct embedd effect on DNA.(6) Restriction enzyme pattern analysis of baicalein and DNA binding sites byelectrophoresis, the study revealed that after effected with baicalein on DNA for druginteraction Taq (T/CGA) restriction site happened obvious change.
