Objectives:To study the inhibitory effect and mechanism of sodium cantharidate (SCA) on HCT116and SW480cell strains of human colorectal cancer.Methods:To cultivate colorectal cancer cells(HCT116cells、SW480cells), interfering with SCA in different concentrations, observing the inhibitory effects SCA has on the proliferation of colorectal cancer cells with the method of MTT, the influence of SCA has on cell apoptosis and cells cycle of colorectal cancer cells by flow cytometry, and expression of Bax、Bcl-2、MLH-1、MSH2and P53after the intervention of SCA with the method of immuno-cytochemistry.Results:SCA had strong inhibitory effect on the growth of HCT116cells and SW480cell strains, and the inhibitory effect on HCT116was stronger than that on SW480cells. SCA could increase apoptosis rate of colorectal cancer cells detected by flow cytometry, and the concentration of SCA and apoptosis rate were in positive correlation. When the concentration of SCA on HCT116reached0.625ug/ml, there is a statistical significance when comparing the experimental group with the control group. When the concentration of SCA on SW480reached1.25ug/ml, there is a statistical significance when comparing the experimental group with the control group. HCT116cells and SW480cells were blocked at G2phase after the intervention of SCA. When the concentration of SCA reached2.5ug/ml, there is a statistical significance when comparing the experimental group with the control group. Immuno-cytochemistry indicated that SCA could increase the expressions of Bax、MLH-1、MSH2、P53of HCT116cells. The concentration of SCA and expressions were in positive correlation, when the concentration of SCA reached0.625ug/ml, the expressions of Bcl-2could be decreased; SCA could increase the expressions of Bax of SW480, but there was no correlation with the concentration of SCA. SCA could increase the expressions of MLH-1、MSH2、p53. The concentration of SCA and expressions were in positive correlation, when the concentration of SCA reached0.625ug/ml, the expressions of Bcl-2could be decreased.Conclusions:SCA had strong inhibitory effect on the growth of HCT116cells and SW480cell strains, and the inhibitory effect on HCT116was stronger than that on SW480cells. As the intervention of SCA could regulate the expression of Bax、Bcl-2、MLH-1、MSH2and p53, HCT116cells and SW480cells have G2phase block and apoptosis, but and the influence on HCT116was stronger than that on SW480cells.
