Objective:We studied the effects of several benefiting qi and invigorating kidney Chinese herb water extract on JAK/STAT signaling pathway and cell proliferation in mice glomerular mesanfial SV40MES13cells.Method:The cytotoxcity of benefiting qi and invigorating kidney Chinese herb water extract on mice glomerular mesanfial SV40MES13cell was determined by MTT assay. Western blot analysis was used to detect the effect of IL-6to STAT3activation and the effects of benefiting qi and invigorating kidney Chinese herb on pSTAT3, STAT3and FN expression in SV40MES13cells.Results:Four benefiting qi and invigorating kidney Chinese herb dodder, Chinese yam, tuckahoe, and codonopsis pilosula water extract are own bidirectional regulatory effects on cell proliferation of SV40MES13cells. They can induce cell proliferation in low concentration and inhibit cell proliferation in high concentration. Dangsen and dogwood only showed inhibitory effects on SV40MES13cell proliferation. However, astragalus and Codonopsis pilosula exhibited more potent inhibition on SV40MES13cells than dodder, Chinese yam and Tuckahoe. The IC50of Dangsen and dogwood extract on SV40MES13cell are similar but dogwood exhibited strongest inhibition on SV40MES13cell proliferation.Low level expression of p-STAT3was detected in SV40MES13cells and IL-6can induce STAT3activation as fast as20min. IL-6treatment induced upreguation of pSTAT3and FN, but dogwood water extract inhibited IL-6-induced STAT3activation and FN expression in a dose-dependent manner in SV40MES13cells.Conclusion:The extract of benefiting qi and invigorating kidney Chinese herb can inhibit glomerular mesanfial cell proliferation in high concentration. STAT3phosphorylation in mice glomerrular mesanfial SV40MES13cell can be induced by IL-6. Dogwood extract inhibited IL-6induced STAT3activation and FN expression. Therefore, the inhibitory effect of proliferation by dogwood could be mediated by inhibiting IL-6/JAK/STAT3pathway and dogwood could be own anti-renal fibrisis effect. |
