ObjectiveEsophageal cancer is one of the most frequently-occurring malignanciesin our country which take serious harm to human health. The purpose of thisstudy was to detect the expression of HDAC6mRNA in esophageal squamouscell carcinoma(ESCC) tissues and adjacent noncancerous tissues,as well as toinvestigate the relation between HDAC6mRNA’s expression and clinicalpathologic characteristics of ESCC.We hope to provide the test data for HDAC6mRNA to become an early clinical diagnostic marker.Methods42groups of ESCC tissues and adjacent noncancerous tissues were selectedwithin30minutes after surgery which were not given radiotherapy andchemotherapy before surgery. The Semi-quantitative RT-PCR method waschosen to investigate the relative expression levels of HDAC6mRNA in ESCCtissues and adjacent noncancerous tissues.We used SPSS17.0version17.0toanalyze HDAC6mRNA’s expression in ESCC tissues and adjacentnoncancerous tissues,as well as to investigate the relation between HDAC6mRNA’s expression and clinical pathologic characteristics of ESCC.Results1. The positive ratio of HDAC6mRNA in ESCC tissues (85.71%,36/42)was significantly higher than that in adjacent noncancerous tissues(66.67%,28/42).(χ~2=4.200P=0.040<0.05).2. The relative expression levels of HDAC6mRNA in ESCC tissues(0.57±0.26) was significantly higher than that in adjacent noncancerous tissues(0.23±0.20).(t=6.745, P=0.00<0.001).3. According to the patient’s gender,the positive ratio of HDAC6mRNA inmale(84.85%) has no statistically significant than that in the femail%(88.89%) (χ~2=0.000, P=1.000>0.05)4. According to the patient’s age,the positive ratio of HDAC6mRNA in theelder group over60years(86.96%) has no statistically significant than that inthe group less than60years(84.21%).(χ~2=0.000, P=1.000>0.05).5. According to the tumor size,the positive ratio of HDAC6mRNA in thegroup which has larger tumor size than5cm(82.76%) has no statisticallysignificant than the other group (92.31%).(χ~2=0.116, P=0.733>0.05).6. According to the invasion depth of tumor,the positive ratio of HDAC6mRNA in ESCC tissues is not related to its invasion depth (χ~2=0.004,P=0.949>0.05).7. According to the histologic grade of tumor,the expression levels ofHDAC6mRNA in ESCC tissues is not related to its histologic grad(e χ~2=0.005,P=0.941>0.05).8. According to lymph node metastasis status,the positive ratio of HDAC6mRNA in lymph node metastasis tumors(100.00%)was significantly higher thanthat in lymph no node metastasis(68.42%)(χ~2=6.091, P=0.014<0.05). Therelative expression levels of HDAC6mRNA in ESCC tissues has positivecorrelation with the degree of lymph node metastasis.9. According to the TNM staging of tumor,the positive ratio of HDAC6mRNA in early-stage (I、 II) was significantly lower than that in late-stage(III,IV)(χ~2=4.331, P=0.037<0.05). The relative expression levels of HDAC6mRNA in ESCC tissues has positive correlation with TNM staging.Conclusion1. The positive ratio and relative levels of HDAC6mRNA in ESCC tissuesis significantly higher than that in the adjacent noncancerous tissues.2. The expression of HDAC6mRNA in ESCC tissues is not related topeople’s sex,age,tissue size,histologie grade and invasion depth.3. The positive ratio and relative levels of HDAC6mRNA in ESCC tissueshas positive correlation with lymph node metastasis.4. The positive ratio and relative levels of HDAC6mRNA in ESCC tissueshas positive correlation with TNM staging.
