Objective:To research the expression of MCP-1in gastric cance、normal gastric tissue、omentum and gastric lymph node and significance.Methods:The specimens of fifty patients with gastric cancer operated in Qingdao Municipal Hospital between March2012and Dec2013including gastric cancer normal gastric tissue、omentum、gastric lymph node,were collected.Besides, we gathered ten specimens of normal omentum. The lymph node were handled and observed by Department of Pathology before the experiment. Enzyme linked immunosorbent assay(ELISA) and reverse transcription-polymerase chain reaction(RT-PCR) were applied to detect the content of MCP-1or MCP-1RNA in gastric cancer、normal gastric tissue and omentum and the content of MCP-1in gastric lymph node was determined by immunohistochemistry.Results:(1) ELISA detected the difference of the content of MCP-1between gastric cancer and normal gastric tissue was significant(P<0.05), and the content of gastric cancer increased with the increase of clinical stage、T stage、N stage and decreased with the increase of the tumor differentiation degree, the difference was statistically significant(F=496.235,325.475,519.614,697.214,P<0.05).The difference between I and Ⅱstage/T1and T2stage、N0and N1stage were not statistically significant(P>0.05). The difference of MCP-1between gastric and normal omentum was significant(P<0.05), while there was no difference between Ⅰ、Ⅱ、T1、T2、N0、N1stage、well-differentiated omentum and normal omentum(P>0.05).The difference in clinical stage、T stage、N stage、difference stage was significant(F=474.529,514.218,498.657,577.452, P<0.05),while the difference between Ⅰ and Ⅱ stage、T1and T2stage、NO and Nl stage in omentum is not significant(P>0.05).(2) RT-PCR detected the content of MCP-1RNA of gastric cancer was higher than normal gastric tissue, and the difference was significant (P<0.05). The content of MCP-1RNA in different clinical stage、T stage、N stage、differentiation degree was statistically significant(F=596.006,612.453,687.554,876.659,P<0.05). The difference between gastric omentum and normal omentum was significant(P<0.05),while the content of MCP-1RNA of Ⅱ、Ⅱ、T1、T2、N0、N1stage、well-differentiated omentum was not different with normal omentum(P>0.05). The difference of the content of MCP-1RNA in different clinical stage、T stage、N stage、differentiation stage was statistically significant(F=519.871,523.769,629.463,617.893, P<0.05).There was no difference between Ⅰ and Ⅱ stage、T1and T2stage、NO and N1stage omentum(P>0.05).(3) Immunohistochemistry detected the positive rate of gastric lymph node was related with clinical stage、T stage、N stage and differentiation stage, which was increased with the increase of clinical stage、T stage、N stage and decrease of differentiation degree, and the difference was significant(x2=144.020,69.605,30.402, P<0.01).Conclusion:The content of MCP-1in gastric cancer、omentum and gastric lymph node was related with clinical stage、T stage、N stage、differentiation degree,which increased with the increase of stage and decrease of differentiation degree, and may reflect the degree of tumor progression.
