Objective: To investigate the association about vascular endothelial growthfactor (VEGF), its receptors(VEGFRs), hypoxia-inducible factor-1α (HIF-1α) andsevere preeclampsia.Methods:Selecting60cases of pregnant women who were not in labor fromobstetrical departments of Jiangxi Maternal and Child Hospital between September2011and June2012, which obtained30cases of severe preeclampsia pregnancy and30cases of normal control pregnancy. Serum levels of VEGF, VEGFR-2, sFlt-1andHIF-1α were detected by enzyme-linked immunosorbent assay, placental contents ofVEGF and VEGFR-2were assessed by immunohistochemistry, and the mRNA levelsof VEGF, VEGFR-2and sFlt-1were assessed by RT-PCR.Results:1. The serum and placenta VEGF expressions were significantly decreased in severepreeclampsia, while the placenta VEGF mRNA levels were significantlyup-regulated.2. The serum and placenta VEGFR-2expressions were significantly decreased insevere preeclampsia, while the placenta VEGFR-2mRNA levels weresignificantly up-regulated.3. The serum sFlt-1levels and placenta sFlt-1mRNA levels were significantlyup-regulated in severe preeclampsia.4. The serum HIF-1α levels were significantly up-regulated in severe preeclampsia.Conclusions:The decreased expressions of VEGF/VEGFR-2in placenta may impactplacental vascular remodeling. The decreased expressions of VEGF/VEGFR-2andthe raised expressions of sFlt-1/HIF-1α in serum may cause maternal severepreeclampsia symptoms. All these factors are involved in the pathogenesis of severepreeclampsia.
