Objective:To investigate the effect of ketamine,propofol exposure in early pregnancy on the expression of hippyragranin (HGN) mRNA in hippocampus of offspring rats’, thus explore part of mechanism of their effects on learning and memory function.Methods:Two hundreds and ten female Sprague-Dawley rats aged7weeks,weighing270-300g,which at their gestation of5-7days were ramdomly divided into7groups (n=30each);2h anesthesia with ketamine group(group K1),4h anesthesia with ketamine group(group K2),8h anesthesia with ketamine group(group K3),2h anesthesia with propofol group(group P1),4h anesthesia with propofol group(group P2),8h anesthesia with propofol group(group P3) and control group(group C).Ketamine20mg/kg was injected via the caudal vein,followed by continuous intravenous infusion at130mg-1.kg-1.h-1, after loss of righting reflex, and the infusion lasted for2,4and8h in group K1-K3, respectively. Propofol20mg/kg was injected via the caudal vein, after loss of righting reflex, followed by continuous intravenous infusion at20mg-1.kg-1.h-1lasted for2,4and8h in group P1-P3, respectively. The equal volume saline was given instead of ketamine or propofol in group C. Morris water maze experiment was performed to evaluate the learning and memory function of offsprings rats’(aged30days).After Morris water maze experiment, the hippocampus of offsprings rats’were collected. Real time polymerase chain reaction was used to determine the expression levels of HGN mRNA in the offspring rats’ hippocampus.Results:(1) The escape latency of the offspring rats’ in group Kl, K2and K3prolonged significantly when compared with these in group C(P<0.05), the target quadrant traveling time and the platform-crossing frequencies of the offspring rats in group K1, K2and K3reduced significantly than group C(P<0.05), though no significant differences could be seen among the later three groups(P>0.05).The expression of HGN mRNA was significantly higher in group K3than that in group K1and group K2(P<0.05), there was no significant difference between group K1and group K2(P>0.05).(2) Compared with group C, the escape latency of the offspring rats’ in group P2and P3prolonged significantly, the target quadrant traveling time and the platform-crossing frequencies reduced significantly(P<0.05),yet propofol anesthesia for2hours(group P1) did not show this effect(P>0.05). The escape latency, the target quadrant traveling time and the platform-crossing frequencies had no differences among group P1, group P2and group P3(P>0.05). The expression levels of HGN mRNA in the offsprings’ hippocampus in group P1and group P2had no statistical differences compared with group C(P>0.05),while the expression of HGN mRNA in group P3was obviously more than that in group C(P<0.05). Compared with group P1, the expression levels of HGN mRNA in the offsprings’ hippocampus in group P3increased significantly(P<0.05), there was no significant difference between group P1and group P2(P>0.05);Compared with group P2,the expression levels of HGN mRNA in group P3increased significantly(P<0.05).Conclusions:Ketamine or propofol exposure during the early pregnancy could damage the learning and memory function of offspring rats’, its mechanism might be associated with the up-regulation of the expression of HGN mRNA in offspring rats’ hippocampus.
